Description:
ZBED1, also known as DREF, is the human homolog of the Drosophila protein DREF, a transcriptional regulatory factor required for expression of genes involved in DNA replication and cell proliferation. RNA interference (RNAi) experiments targeting ZBED1 RNA resulted in the inhibition of S phase entry and reduction of histone H1 mRNA in HeLa cells, suggesting that ZBED1 also plays a role in cell proliferation in human cells. At least 22 ribosomal proteins (RPs) possess ZBED1 binding sites in their promoters; anti-ZBED1 RNAi also resulted in decreased RP gene expression. Immunoprecipitation analyses have shown that ZBED1 self-associates in vivo via a carboxyl-terminal hATC domain, and this self-association is required for nuclear localization and DNA binding.

Alpha-tubulin belongs to the tubulin superfamily, which is composed of six distinct families. Along with beta-tubulins, alpha-tubulins are the major components of microtubules. These microtubules are involved in a wide variety of cellular activities ranging from mitosis and transport events to cell movement and the maintenance of cell shape. Alpha- and beta-tubulin dimers are assembled to 13 protofilaments that form a microtubule of 22-nm diameter. Tyrosine ligase adds a C-terminal tyrosine to monomeric alpha-tubulin. Assembled microtubules can again be detyrosinated by a cytoskeleton-associated carboxypeptidase. Another post-translational modification of detyrosinated alpha-tubulin is C-terminal polyglutamylation, which is characteristic of microtubules in neuronal cells and the mitotic spindle. Like GAPDH and beta-Actin, this antibody makes an excellent loading control in immunoblots.

Regulator of G-protein signaling (RGS) proteins contain an 120 amino acid conserved domain, termed the RGS domain, that acts as a GTPase-activating protein that acts to reduce the signal transmitted by the receptor-activated G-alpha subunit. RGS22 is a recently identified member of this family that localizes to the testis and can interact with guanine nucleotide binding proteins alpha 11, 12, and 13 (GNA11, GNA12, and GNA13). While RGS22 has been postulated to play a role in spermiogenesis in the testis, it is also expressed in several cancer cell lines with an epithelial origin and associated with cancer metastasis. Its overexpression in a highly metastatic cancer causes a decrease in cell migration and a reduction of the invasive potential of the cells, suggesting that RGS22 may be a potential prognostic biomarker for metastasis.

FGF13is a large gene, extending over approximately 200 kb in Xq26.3, and contains at least 7 exons. By cytogenetic, FISH, and database analysis, Gecz et al. (1999) localized the FGF13 gene within a 400-kb duplication interval on chromosome Xq26.3. FGF family members possess broad mitogenic and cell survival activities, and are involved in a variety of biological processes, including embryonic development, cell growth, morphogenesis, tissue repair, tumor growth, and invasion. Other areas of interests and use for researchers include cancer, borjeson-forssman-lehmann syndrome, regulation of actin cytoskeleton pathways, apoptosis, GPCR pathways, TGF-Beta pathways, and Rho familty GTPase pathways. This gene is located to a region associated with Borjeson-Forssman-Lehmann syndrome (BFLS), a syndromal X-linked mental retardation, which suggests it may be a candidate gene for familial cases of the BFL syndrome.
