The RM4-5 monoclonal antibody specifically reacts with mouse CD4, also known as L3T4, a 55 kDa differentiation antigen expressed by the majority of thymocytes, subpopulations of mature T cells (like major histocompatibility complex class II-restricted T lymphocytes), a subset of natural killer T cells, and on pluripotent hematopoietic stem cells. CD4 binds to the major histocompatibility complex class II (MHC class II) and enhances T lymphocyte development and mature T cells functions. In T lymphocytes, CD4 binds to the cytoplasmic tail of enzyme tyrosine kinase (p56lck).Binding of RM4-5 is blocked by the anti-mouse CD4 clone GK1.5.

The 145-2C11 monoclonal antibody reacts with mouse CD3e, the 20 kDa ε chain of the TCR complex. Together with the γ and δ subunits of CD3, the ε subunits are involved in the assembly, trafficking, and surface expression of T-cell receptor complex. CD3 is expressed on thymocytes, mature T cells, and natural killer T cells, and the ε chain enhances the antigen recognition.The 145-2C11 antibody binds to the TCR complex and, depending on the conditions, initiates T cells activation, proliferation, and apoptosis. The soluble antibody seems to block lysis of target cells by antigen-specific cytotoxic T lymphocytes. The 145-2C11 antibody does not cross-react with the rat leukocytes, and it is used as a phenotypic marker for mouse T lymphocytes.

This antibody recognizes a protein of 55-62kDa, identified as Cyclin B1. In mammals, Cyclin B1 associates with inactive p34cdc2, which facilitates phosphorylation of p34cdc2 at amino acids Thr-14 and Tyr-15. This maintains the inactive state until the end of G2-phase. The inactive Cyclin B1-p34cdc2 complex continues to accumulate in the cytoplasm until the completion of DNA synthesis, when Cdc25, a specific protein phosphatase, dephosphorylates amino acids Thr-14 and Tyr-15 of p34cdc2 rendering the complex active at the G2/M boundary. This mitotic kinase complex remains active until the metaphase/anaphase transition when Cyclin B1 is degraded. This degradation process is ubiquitin-dependent and is necessary for the cell to exit mitosis. Cyclin B1-p34cdc2 plays a critical role in G2 to M transition.

MRE11 Antibody: MRE11 is involved in the repair of DNA double strand breaks as part of a complex that includes the Rad50 and NBS1 protein and is thought to act in the same pathway as the A-T mutated (ATM) protein. By itself, the protein has 3' to 5' exonuclease activity and endonuclease activity. The protein forms a complex with the RAD50 homolog; this complex is required for non-homologous joining of DNA ends and possesses increased single-stranded DNA endonuclease and 3' to 5' exonuclease activities. In conjunction with a DNA ligase, this protein promotes the joining of noncomplementary ends in vitro using short homologies near the ends of the DNA fragments. The hexameric protein complex formed by the MCM proteins is a key component of the pre-replication complex (pre_RC) and may be involved in the formation of replication forks and in the recruitment of other DNA replication related proteins.
