Advanced BioMatrix offers PhotoCol®, a purified methacrylated Type I bovine collagen kit. PhotoCol® provides native-like 3D collagen gels with the unique attributes to be prepared at various concentrations and crosslinked to provide various gel stiffness.
The PhotoCol® kit consists of purified methacrylated Type I bovine collagen as the core component with other support reagents in the kit. The methacrylated Type I collagen is produced from telo-peptide intact bovine collagen where the collagen has been modified by reacting the free amines, primarily the e-amines groups of the lysine residues as well as the a-amines groups on the N-termini. Approximately 40% of the total lysine residues of the collagen molecule have been methacrylated. The collagen is extracted from bovine hide and contains a high monomer content. The collagen starting material was isolated from a closed herd and purified using controlled manufacturing processes. A bottle of 20 mM acetic acid solution is provided to solubilize the lyophilized methacrylated collagen at concentrations ranging from 3 to 8 mg/ml.The neutralization solution consists of an alkaline 10X phosphate buffered saline (PBS) solution which provides physiological salts and pH in the final mixture.The photoinitiator consists of Irgacure 2959 to be formulated in methanol (methanol not included) which allows UV crosslinking of the collagen at 365 nm.

The human insulin receptor is a heterotetrameric membrane glycoprotein consisting of disulfide linked subunits in a beta-alpha-alpha-beta configuration. The beta subunit (95 kDa) possesses a single transmembrane domain, whereas the alpha subunit (135 kDa) is completely extracellular. The insulin receptor exhibits receptor tyrosine kinase (RTK) activity. RTKs are single pass transmembrane receptors that possess intrinsic cytoplasmic enzymatic activity, catalyzing the transfer of the gamma phosphate of ATP to tyrosine residues in protein substrates. RTKs are essential components of signal transduction pathways that affect cell proliferation, differentiation, migration and metabolism.Included in this large protein family are the insulin receptor and the receptors for growth factors such as epidermal growth factor, fibroblast growth factor and vascular endothelial growth factor. Receptor activation occurs through ligand binding, which facilitates receptor dimerization and autophosphorylation of specific tyrosine residues in the cytoplasmic portion. The interaction of insulin with the alpha subunit of the insulin receptor activates the protein tyrosine kinase of the beta subunit, which then undergoes an autophosphorylation that increases its tyrosine kinase activity. Three adapter proteins, IRS1, IRS2 and Shc, become phosphorylated on tyrosine residues following insulin receptor activation. These three phosphorylated proteins then interact with SH2 domain containing signaling proteins.

The total protein is measured using the Biuret procedure, with bovine albumin as a standard. Antibody titer of the IgG fraction is standardized by equivalence-point precipitation. F(ab')2 fragments are tested for purity and specificity at a minimum of 20 mg/mL using immunoelectrophoresis. The product is goat F(ab')2; no trace of albumin, whole molecule IgG or Fc is detected.
The F(ab')2 fragment is prepared from the specific goat IgG fraction by limited digestion with pepsin, and purified by gel filtration or affinity chromatography. The resulting F(ab')2 fragment is dialyzed into 0.02M sodium phosphate, 0.14M sodium chloride, pH 7.3, 0.05% sodium azide, adjusted to standard titer, filtered through a 0.22 μm filter, vialed and lyophilized. Goat F(AB')2 fragment to Human IgM is used as a precipitating antibody. It gives sharp precipitin lines in immunoelectrophoresis and immunodiffusion techniques. This product is suitable for conjugation with enzymes, radiolabels, fluorochromes or other markers, and for attachment to solid supports for use as an immunoadsorbant. This product is suitable for use as a primary reagent in enzyme immunoassays, fluorescein immunoassays and blot, cell or tissue immunostaining. F(ab')2 fragments are recommended for staining of cells or tissues which contain Fc receptors. (Note: Affinity purified antibodies are recommended to avoid non-specific binding from the inherent antibodies of host animals).

Foxp3 is expressed by the Treg lymphocytes, whose development and function are influenced by the forkhead protein.
