Description:   A sensitive reagent for measuring the free sulfhydryl content in proteins, peptides, and tissues. Used to characterize reactive thiol groups and photometric determination of thiols and for measuring low-molecular mass thiols such as glutathione in both pure solutions and biological samples, such as blood. It can also measure the number of thiol groups on proteins. Through reaction with aliphatic thiol groups a mixed disulfide of protein thiol and one mole of 2-nitro-5-thiobenzoate per mole of protein sulfhydryl group is being formed. DTNB has little absorbance. Reaction with -SH groups on proteins (from any solvent accessible Cys) under mild alkaline conditions (pH 7-8) produces the 2-nitro-5-thiobenzoate anion, which gives an intense yellow color with an absorption maximum at 409.5nm (Extinction coefficient: 14150 M-1*cm-1). Sensitive to various buffer ions, therefore, the extinction coefficient used to calculate the number of sulfhydryl groups must be matched to the reaction conditions. In case the thiol groups are in disulfide bonds, they must be reduced under anaerobic conditions prior to reaction with DTNB.

Description:   AT7519 is a potent inhibitor of several CDK family members. AT7519 showed potent antiproliferative activity (40-940nM) in a panel of human tumor cell lines, and the mechanism of action was shown here to be consistent with the inhibition of CDK1 and CDK2 in solid tumor cell lines. AT7519 caused cell cycle arrest followed by apoptosis in human tumor cells and inhibited tumor growth in human tumor xenograft models. Tumor regression was observed following twice daily dosing of AT7519 in the HCT116 and HT29 colon cancer xenograft models. Also it has been shown that the biological effects are linked to inhibition of CDKs in vivo and that AT7519 induces tumor cell apoptosis in these xenograft models. Moreover, AT7519 has an attractive biological profile and is well tolerated and effective making it a more plausible candidate for clinical development than previously available CDK inhibitors. Description:   Phenol red is an acid-base indicator. It is made by condensing two moles of phenol with one mole of o-sulfobenzoic acid anhydride.
Phenol Red is used as a pH indicator in cell culture applications. A solution of phenol red will have a yellow color at a pH of 6.4 or below and a red color at a pH of 8.2 and above. Phenol red in tissue culture media can act as a weak estrogen, especially with human breast cancer cells.
