Description:   mTOR controls cell growth, in part by regulating p70 S6 kinase alpha (p70alpha) and eukaryotic initiation factor 4E binding protein 1 (4EBP1). Raptor is a 150 kDa mTOR binding protein that also binds 4EBP1 and p70alpha. The binding of Raptor to mTOR is necessary for the mTOR-catalyzed phosphorylation of 4EBP1 in vitro, and it strongly enhances the mTOR kinase activity toward p70alpha. Rapamycin or amino acid withdrawal increases, whereas insulin strongly inhibits, the recovery of 4EBP1 and raptor on 7-methyl-GTP Sepharose. Partial inhibition of raptor expression by RNA interference (RNAi) reduces mTOR-catalyzed 4EBP1 phosphorylation in vitro. RNAi of C. elegans raptor yields an array of phenotypes that closely resemble those produced by inactivation of Ce-TOR. Thus, raptor is an essential scaffold for the mTOR-catalyzed phosphorylation of 4EBP1 and mediates TOR action in vivo.
Supplier:  Adipogen
Description:   Catabolite of thymine. Precursor for 3-Hydroxyisobutyric acid synthesis. Small molecule myokine. Browning inducer of white adipose tissue (WAT) into brown adipose tissue (BAT). PGC-1alpha-mediated and exercise-triggered nonadrenergic activator of the thermogenic program in WAT. Increases the expression of brown adipocyte-specific genes (browning) in white adipocytes and beta-oxidation in hepatocytes through a PPARalpha-mediated mechanism. Induces a brown adipose-like phenotype in human pluripotent stem cells and improves glucose homeostasis. Enhances fatty acid oxidation and reduces body weight in mice through an increased production of leptin by the white adipose tissue (WAT). Inhibits Agxt2-mediated metabolism of dimethylarginines. Partial agonist of the glycine receptor (GlyR).

Description:   Members of the BAGE gene family encode antigens that are recognized by cytotoxic T lymphocytes and are also known as CT (cancer/testis) antigens. Generated by juxtacentromeric shuffling of the MLL3 gene, the ancestral BAGE gene was expanded by acrocentric exchanges and/or juxtacentromeric movements.Generally, BAGE proteins are silent in all normal tissues with the exception of testis. BAGE2 and BAGE 3 (B melanoma antigen 2 and 3, respectively), also known as Cancer/testis antigen 2.2 and 2.3 (respectively), are 109 amino acid secreted proteins that are expressed in 22% of melanomas, lung and bladder carcinomas, and are also expressed in normal testis tissue. Like the genes encoding MAGE proteins, BAGE genes are most likely silenced by DNA methylation and/or chromatin compaction in normal tissues other than testis.

Description:   This antibody recognizes a protein of HMW, identified as mucin 3 glycoprotein (MUC3). Its epitope localizes between aa SITTTE. This MAb shows no cross-reaction with human milk fat globule membranes, MUC1, or MUC2. MUC3 is distributed in colon and rectum, and is also present to a lesser extent in breast, lung and salivary gland tissues. The Mucins are a family of highly glycosylated, secreted proteins with a basic structure consisting of a variable number of tandem repeats (VNTRs) encoded by 60 base pairs (Mucin 1), 69 base pairs (Mucin 2) and 51 base pairs (Mucin 3). The number of repeats is highly polymorphic and varies among different alleles. Mucin 1 proteins are expressed as type I membrane proteins in addition to secreted forms. Mucin 1 is aberrantly expressed in epithelial tumors including breast carcinomas. Mucin 2 coats the epithelia of the intestines and airways and is associated with colonic tumors. Mucin 3 is a major component of various mucus gels and is broadly expressed in normal and tumor cells.

Description:   The Polycomb group (PcG) genes contribute to the maintenance of cell identity, cell cycle regulation and oncogenesis. SUZ12 (suppressor of zeste 12) is a 90 - 95 kDa member of the polycomb group of transcriptional regulators. It is part of a transcription repression complex termed PRC2 (Polycomb repressive complex 2) that methylates histones in the nucleus, resulting in homeotic (pattern-inducing) gene silencing. It contains one C2H2-type zinc finger region and a C-terminal VEFS-box. SUZ12 is overexpressed in breast and colon cancer. SUZ12 may be a cause of endometrial stromal tumors.

Description:   The Ubiquitin-like containing PHD and RING finger domains 1-binding protein 1 (UHRF1BP1), also known as ICBP90, is a transcription and cell cycle regulator. It specifically binds to the histone H3 N-terminal tail when methylated on K9 by two functional domains, a PHD finger that defines the binding specificity and an SRA (SET- and RING-associated) domain that promotes binding activity. UHRF1BP1 is required for proper heterochromatin formation in mammalian cells. Furthermore, UHRF1BP1 is thought to be a pivotal target for the ERK1/2 signaling pathway to control the proliferation of Jurkat T cells.

Description:   EZH2 was initially identified as a homolog of the drosophila Enhancer of Zeste through exon trap screening of chromosome 21. Both EZH2 and the related protein EZH1 can form complexes with the noncanonical Polycomb repressive complex-2 (PRC2) and maintain repressive chromatin, but the PRC2-EZH1 complex mediates methylation of histone H3. Both EZH1 and EZH2 are thought to function in the maintenance of embryonic stem cell pluripotency and plasticity and recently have been shown to be essential for hair follicle homeostasis and wound repair. Overexpression of EZH2 has been reported as a marker for advanced and metastatic cancers.

Description:   Maelstrom Antibody: The mammalian homolog of the Drosophila protein Maelstrom is expressed in the male germline and localizes to the sex body in spermatocytes and the chromatoid body in round spermatids. Similar to its expression in Drosophila, Maelstrom is a component of nuages, a germ-cell specific organelle and is thought to be essential for spermatogenesis and transposon repression during meiosis. In humans, Maelstrom has been found to be expressed only in the testis and in various cancer cell lines. Treatment of these cell lines with the demethylating agent 5'-Aza-2-Deoxycytidine significantly upregulated Maelstrom levels, indicating that its expression is regulated by DNA methylation.

Description:   WD40 repeats are a common structural module in eukaryotic proteins, and proteins containing WD40 domains have a wide range of functions, including signal transduction, cell cycle regulation, RNA splicing, and transcription. One such protein, WDR35, also known as CED2, has been shown to be mutated in patients with Sensenbrenner syndrome/cranioectodermal dysplasia (CED), an autosomal-recessive disease that is characterized by craniosynstosis and ectodermal and skeletal abnormalities. WDR35 localizes to cilia and dentrosomes during embryogenesis and human and mouse fibroblasts that lack this gene fail to produce cilia. Mutations in this gene can also cause short-rib polydactyly syndromes due to abnormal ciliogenesis.

Description:   The N-methyl-D-aspartate receptor 2B, also names as GRIN2B. The sequence of the predicted 1,484-amino acid human protein is 98% and 96% identical to the sequences of the rat and mouse Nmdar2b proteins, respectively. Nmdar2B gene is located on mouse chromosome 6 between Rho and Ly49 centromerically and Glb telomerically. Mapping of the human NMDAR2B receptor subunit gene (GRIN2B) to chromosome 12p12 overexpression of NMDA receptor 2B (NR2B) in the forebrains of transgenic mice leads to enhanced activation of NMDA receptors, facilitating synaptic potentiation in response to stimulation at 10-100 Hz. This antibody is suitable for researchers interested in neuroscience research.

Description:   Glutamate receptors are the predominant excitatory neurotransmitter receptors in the mammalian brain and are activated in a variety of normal neurophysiologic processes. These receptors are heteromeric protein complexes composed of multiple subunits, arranged to form ligand-gated ion channels. The classification of glutamate receptors is based on their activation by different pharmacologic agonists. The subunit encoded by this gene belongs to a family of AMPA (alpha-amino-3-hydroxy-5-methyl-4-isoxazole propionate)-sensitive glutamate receptors, and is subject to RNA editing (AGA->GGA; R->G). Alternative splicing of this gene results in transcript variants encoding different isoforms, which may vary in their signal transduction properties.

Description:   Chromatin condensation and nuclear fragmentation (CCNF) is the hallmark of apoptosis. CCNF is triggered by the activation of members of caspase family, caspase activated DNase (CAD/DFF40), and several novel proteins including AIF and CIDE. A new inducer of chromatin condensation was recently identified and designated Acinus (for apoptotic chromatin condensation inducer in the nucleus). Acinus is cleaved by caspase-3 and an additional unknown protease generating a small active peptide p17, which causes chromatin condensation in vitro when it is added to purified nuclei. Acinus also induces apoptotic chromatin condensation in cells. Acinus is ubiquitously expressed. Three different spliced forms of Acinus have been identified in human and mouse and designated AcinusL, AcinusS and AcinusS'.

Description:   The DPF3 protein, also known as Cerd4, is a member of the d4 gene family of transcription modulators that also includes DPF1/Neud4 and DPF2/Requiem. DPF3 has been shown to be a epigenetic key factor for heart and muscle development and can bind to methylated and acetylated lysine residues of histone 3 and 4, suggesting that DPF3 may play a role in recruiting chromatin remodeling complexes to acetylated histones. Two isoforms of DPF3, DPF3a and DPF3b, are required as transcriptional co-activators in SWI/SNF complex-dependent activation of the NF-kappaB RelA/p50 heterodimer.
