Description:   Involved in several stages of intracellular trafficking. Interacts with membranes containing phosphatidylinositol 3-phosphate (PtdIns(3P)) or phosphatidylinositol 3,5-bisphosphate (PtdIns(3,5)P2) (PubMed:12198132). Acts in part as component of the retromer membrane-deforming SNX-BAR subcomplex. The SNX-BAR retromer mediates retrograde transport of cargo proteins from endosomes to the trans-Golgi network (TGN) and is involved in endosome-to-plasma membrane transport for cargo protein recycling. The SNX-BAR subcomplex functions to deform the donor membrane into a tubular profile called endosome-to-TGN transport carrier (ETC) (Probable). Can sense membrane curvature and has in vitro vesicle-to-membrane remodeling activity (PubMed:19816406, PubMed:23085988). Involved in retrograde endosome-to-TGN transport of lysosomal enzyme receptors (IGF2R, M6PR and SORT1) and Shiginella dysenteria toxin stxB. Plays a role in targeting ligand-activated EGFR to the lysosomes for degradation after endocytosis from the cell surface and release from the Golgi (PubMed:12198132, PubMed:15498486, PubMed:17550970, PubMed:17101778, PubMed:18088323, PubMed:21040701). Involvement in retromer-independent endocytic trafficking of P2RY1 and lysosomal degradation of protease-activated receptor-1/F2R (PubMed:16407403, PubMed:20070609). Promotes KALRN- and RHOG-dependent but retromer-independent membrane remodeling such as lamellipodium formation; the function is dependent on GEF activity of KALRN (PubMed:20604901). Required for endocytosis of DRD5 upon agonist stimulation but not for basal receptor trafficking (PubMed:23152498).

Description:   The octapeptide circulatory hormone Angiotensin II is a critical regulator of cardiovascular homeostasis formed in blood, with dual function in tissues of the brain, kidney, heart, and vasculature as an autocrine/paracrine growth hormone. The actions of angiotensin II are mediated by type one (AT1) and type two (AT2) angiotensin II receptors, members of the seven-transmembrane Gprotein coupled receptor (GPCR) family. AT1 receptors couple predominantly with Gq/11 alpha subunits, and signal through phospholipases C, D, and A2, inositol phosphates, and calcium channels, as well as a variety of serine/threonine and tyrosine kinases. AT1 expression is greatest in vascular smooth muscle, liver, kidney, adrenal cortex, brain, and lung, and is primarily responsible for the physiological effects of angiotensin II, including regulation of arterial blood pressure, electrolyte and water balance, thirst, hormone secretion, and renal function. The AT2 receptor displays approximately 30% identity to the AT1 receptor, and is highly expressed during embryonic development. AT2 expression in adult tissue is limited to the adrenal medulla, brain, and reproductive tissues, but is upregulated in injured tissue following myocardial infarction, vascular injury, and wound healing. AT2 receptor signaling is not dependent on G-protein coupling or receptor phosphorylation, although coupling to Gia2 and Gia3 alpha subunits occurs in some tissues. AT2 activation can stimulate tyrosine and serine/threonine phosphatases through activation of nitric oxide and cGMP, and appears to counteract the stimulatory effects of AT1 receptor and growth factor receptor signaling.

Description:   The RM4-5 monoclonal antibody specifically reacts with mouse CD4, also known as L3T4, a 55 kDa differentiation antigen expressed by the majority of thymocytes, subpopulations of mature T cells (like major histocompatibility complex class II-restricted T lymphocytes), a subset of natural killer T cells, and on pluripotent hematopoietic stem cells. CD4 binds to the major histocompatibility complex class II (MHC class II) and enhances T lymphocyte development and mature T cells functions. In T lymphocytes, CD4 binds to the cytoplasmic tail of enzyme tyrosine kinase (p56lck).Binding of RM4-5 is blocked by the anti-mouse CD4 clone GK1.5.

Description:   The MEL-14 monoclonal antibody specifically reacts with L- selectin (CD62L), a receptor with lectin-like and Epidermal Growth Factor-like domains. The weight of the CD62L molecules depend on their origin: 74 kDa (on lymphocytes) or 95 kDa (on neutrophils). In the Mouseorganism, CD62L can be expressed by most thymocytes, on subsets of B and T lymphocytes, monocytes, eosinophils, and neutrophils. The L-selectin binds sulfated, fucosylated, and glycosylated glycoproteins (MadCAM-1, GLYCAM-1, and CD 34). It mediates the migration of lymphocytes to the site of inflammation and their return to the peripheral lymphoid tissues and to the HEV (high endothelial venules). In vitro, L-selectin inhibits this binding and the lymphocyte extravasation into peripheral lymph nodes.

Description:   The 145-2C11 monoclonal antibody reacts with mouse CD3e, the 20 kDa ε chain of the TCR complex. Together with the γ and δ subunits of CD3, the ε subunits are involved in the assembly, trafficking, and surface expression of T-cell receptor complex. CD3 is expressed on thymocytes, mature T cells, and natural killer T cells, and the ε chain enhances the antigen recognition. The 145-2C11 antibody binds to the TCR complex and, depending on the conditions, initiates T cells activation, proliferation, and apoptosis. The soluble antibody seems to block lysis of target cells by antigen-specific cytotoxic T lymphocytes. The 145-2C11 antibody does not cross-react with the rat leukocytes, and it is used as a phenotypic marker for mouse T lymphocytes.
