Immunoglobulin G (IgG) is a glycoprotein antibody that regulates immune responses such as phagocytosis and is also involved in the development of autoimmune diseases. Mouse IgGs have four distinct isotypes, namely IgG1, IgG2a, IgG2b, and IgG3. IgG1 regulates complement fixation in mice. Anti-Mouse IgG (whole molecule)-FITC antibody reacts with normal mouse serum and all mouse IgGs. Fluorescein-Conjugated Goat Affinity Purified Antibody to mouse IgG (whole molecule) is used in immunostaining of acetone-fixed frozen and formalin-fixed, paraffin-embedded tissue sections. It is suitable for use as a reagent in immunofluorescence assays (IFA), cell staining (fluorescent microscopy and cell sorting), tissue staining, and blot immunostaining. Use of these affinity purified antibodies is recommended to avoid non-specific binding from the inherent antibodies of host animals. (Note: F(ab')2 fragments are recommended for staining of cells or tissues which contain Fc receptors). Antibody and FITC are conjugated under defined conditions to obtain the specified molar ratio (3-6 mol/mol). Conjugated protein is purified by gel filtration. The product is dialyzed into 0.02M sodium phosphate, 0.14M sodium chloride, pH 7.3, with 0.05% sodium azide and 10% glycerol (v/v), adjusted to standard titer, filtered through a 0.22 μm filter, and vialed. The total protein is measured by absorbance (A280 nm); F/P ratio is calculated using absorbance at 491.5 nm. Antibody titer of the affinity purified antibody is standardized by equivalence-point precipitation. The affinity purified antibody is tested for purity at 10 mg/mL using immunoelectrophoresis; No trace of albumin is detected. Antibody activity to non-immunoglobulin serum proteins is not present.

Description:
The GK1.5 monoclonal antibody specifically binds with the mouse CD4 molecule, also known as L3T4, a 55 kDa differentiation antigen which binds to the MHC class II. CD4 is expressed on most thymocytes, a subpopulation of mature T lymphocytes, dendritic cells, pluripotent hematopoietic stem cells, B cell precursors, and lymphoid precursors inside the thymus. It is also expressed on the mouse egg cell membrane, enhancing adhesion to MHC class II bearing sperm. By interaction with MHC class II on the surface of APC, CD4 initiates the development of T lymphocytes and helps the optimum functioning of mature T lymphocytes. The binding of the GK1.5 antibody blocks the binding of the Anti-Mouse CD4 RM4-5 antibody.

The M5/114.15.2 monoclonal antibody or (M5/114) is specific for the mouse major histocompatibility complex class II, specific for the I-A and I-E subregion-encoded glycoproteins (alloantigens I-Ab, I-Ad, I-Aq, I-Ed, and I-Ek, but not the I-Af, I-Ak, nor I-As antigens). The antibody also reacts with cells from the H-2p or H-2r haplotypes mice. The polymorphic determinant represented by the mouse major histocompatibility complex class II (MHC Class II) is expressed by the immune cells bearing the specified alloantigens, like T and B lymphocytes, monocytes, dendritic cells, and macrophages. The 16-10A1 antibody reacts with mouse CD80, also known as B7-1, a 55 kDa type I transmembrane protein ligand for CD152 (CTLA-4) and for CD28, a co-stimulatory receptor for the T cell receptor (TCR). CD28 also binds a second B7 ligand known as CD86 (B7-2). Both CD80 and CD86 are expressed on activated B cells and antigen-presenting cells. These ligands trigger CD28 signaling in concert with TCR activation to drive T cell proliferation, induce high-level expression of IL-2, impart resistance to apoptosis, and enhance T cell cytotoxicity.
