Description:
Adducins are a family of cytoskeleton proteins encoded by three genes (alpha, beta, gamma). Adducin is a protein associated with the inner leaflet of the plasma membrane and is one of the proteins localized at the spectrin-actin junction of the membrane skeleton. The cortical actin cytoskeletal network is lost during apoptosis and Adducins are central in the cortical actin network organization. Adducin Alpha is a cytoskeletal protein involved with sodium-pump activity in the renal tubule and is associated with hypertension. The expression of Adducin Alpha and Adducin Gamma is ubiquitous in contrast to the restricted expression of Adducin Beta. Adducin Beta is expressed at high levels in brain and hematopoietic tissues, such as bone marrow, in humans, and in spleen in mice.

BAMBI is a membrane spanning glycoprotein that acts as a negative regulator of TGF-beta signaling during development. The BAMBI family of proteins are related to type I TGF-beta receptor family, however, BAMBI is a pseudoreceptor that lacks an intracellular serine/threonine kinase domain. BAMBI transcription regulation is under the influence of beta-catenin, BMP, smad3 and smad4. BAMBI expression can increase in colorectal and hepatocellular carcinomas relative to non-cancerous tissues. BAMBI is expressed at high levels during odontogenesis. BAMBI is coexpressed with Bmp-4 during early Xenopus embryogenesis and can be detected in poorly metastatic human melanoma cell lines.

In general, the protein phosphatase (PP) holoenzyme is a trimeric complex composed of a regulatory subunit, a variable subunit, and a catalytic subunit. Four major families of protein phosphatase catalytic subunits have been identified, designated PP1, PP2A, PP2B (calcineurin) and PP2C. An additional protein phosphatase catalytic subunit, PPX (also known as PP4) is a putative member of a novel PP family. The PP2A family comprises subfamily members PP2A Alpha and PP2A Beta. The PP2A catalytic subunit associates with a variety of regulatory subunits. Regulatory subunits include PP2A-A-Alpha and -A-Beta, PP2A-B-Alpha and -B-Beta, PP2A-C-Alpha and -C-Beta, PP2A-B56-Alpha, -B56-Beta, -B56-gamma and -B56-Delta.

Cadherins are calcium-dependent cell-cell adhesion molecules, and protocadherins constitute a subfamily of nonclassic cadherins. Protocadherin beta 13 is a member of the protocadherin beta gene cluster, one of three related gene clusters tandemly linked on chromosome five. The gene clusters demonstrate an unusual genomic organization similar to that of B-cell and T-cell receptor gene clusters. The beta cluster contains 16 genes and 3 pseudogenes, each encoding 6 extracellular cadherin domains and a cytoplasmic tail that deviates from others in the cadherin superfamily. The extracellular domains interact in a homophilic manner to specify differential cell-cell connections. Unlike the alpha and gamma clusters, the transcripts from these genes are made up of only one large exon, not sharing common 3' exons as expected. These neural cadherin-like cell adhesion proteins are integral plasma membrane proteins. Their specific functions are unknown but they most likely play a critical role in the establishment and function of specific cell-cell neural connections.
