Alpha actinin is an actin-binding protein with multiple roles in different cell types. In nonmuscle cells, the cytoskeletal isoform is found along microfilament bundles and adherens-type junctions, where it is involved in binding actin to the membrane. In contrast, skeletal, cardiac, and smooth muscle isoforms are localized to the Z-disc and analogous dense bodies, where they help anchor the myofibrillar actin filaments. Alpha actinins belong to the spectrin gene superfamily which represents a diverse group of cytoskeletal proteins, including the alpha and beta spectrins and dystrophins. The enzyme phosphatidylinositol 3 kinase (PI3 kinase) is a lipid kinase that generates phosphatidylinositol 3, 4, 5-triphosphate in response to receptor activation in many signal transduction pathways. Class IA PI3Ks exist as a heterodimer of a catalytic 110 kDa (p110) and a regulatory p85 subunit (e.g. p85 alpha). p85 alpha is an adaptor molecule that regulates the activity of the catalytic p110 subunit by binding to phosphorylated receptor tyrosine kinases (RTKs) through its SH2 domain and mediating the interaction between p110 and the plasma membrane. p85 alpha is necessary for insulin-stimulated increase in glucose uptake and glycogen synthesis in insulin-sensitive tissues.

The OKT8 antibody is specific for the 32-34 kDa alpha chain of human CD8, known as CD8a or CD8 alpha. CD8a can form a homodimer (CD8 alpha-alpha), but is more commonly expressed as a heterodimer with a second chain known as CD8b or CD8 beta.
