17-AAG (17-Allylamino-17-demethoxygeldanamycin) is a synthetic derivative of geldanamycin. Like geldanamycin, 17-AAG is recognised for its role as an inhibitor of heat shock protein 90 (Hsp90), a molecular chaperone involved in protein folding and stabilisation.

It is often used as a tool to study Hsp90-dependent processes. By inhibiting Hsp90, 17-AAG impacts the stability and function of various client proteins, many of which are involved in critical cellular processes, including signal transduction and cell cycle regulation. Similar to geldanamycin, 17-AAG is particularly relevant in cancer research, as Hsp90 is often crucial for the stability of oncoproteins. Researchers use 17-AAG to explore the molecular mechanisms of Hsp90 inhibition, with potential applications in understanding cancer biology and developing therapeutic strategies.
