Description:
Binds to DNA, at nuclear matrix- or scaffold-associated regions. Thought to recognize the sugar-phosphate structure of double-stranded DNA. Transcription factor controlling nuclear gene expression, by binding to matrix attachment regions (MARs) of DNA and inducing a local chromatin-loop remodeling. Acts as a docking site for several chromatin remodeling enzymes and also by recruiting corepressors (HDACs) or coactivators (HATs) directly to promoters and enhancers. Required for the initiation of the upper-layer neurons (UL1) specific genetic program and for the inactivation of deep-layer neurons (DL) and UL2 specific genes, probably by modulating BCL11B expression. Repressor of Ctip2 and regulatory determinant of corticocortical connections in the developing cerebral cortex. May play an important role in palate formation. Acts as a molecular node in a transcriptional network regulating skeletal development and osteoblast differentiation.

Description:
The homeobox DNA-binding domain is a 60 amino acid motif that is conserved among many species and functions to bind DNA via a helix-turn-helix structure, thereby playing a role in transcriptional regulation and in the control of gene expression. MIXL1 (Mix1 homeobox-like 1), also known as MIXL, is a 232 amino acid protein that localizes to the nucleus and contains one homeobox DNA-binding domain. Expressed in lymph tissues, MIXL1 functions as a transcription factor that plays an essential role in axial mesendoderm morphogenesis and endoderm formation and is also required for cellular differentiation during blood development. Additionally, MIXL1 is involved in maturation of heart and gut tissue during embryogenesis and may also act as a negative regulator of brachyury expression. Overexpression of MIXL1 is associated with non-Hodgkin and Hodgkin lymphomas, suggesting a role in carcinogenesis.

Description:
Apolipoproteins are protein components of plasma lipoproteins. ApoM (Apolipoprotein M), also known as protein G3a, is a member of the Lipocalin family of proteins. ApoM is exclusively expressed in kidney tubular epithelial cells and liver hepatocytes. Mature ApoM retains its signal peptide, which acts as a hydrophobic anchor, and contains a structurally conserved eight stranded antiparallel ∫ barrel which binds retinol and retinoic acid. ApoM may play a key role in reverse cholesterol transport. It mainly associates with high density lipoprotein (HDL) and to a lesser extent with triglyceride-rich lipoprotein (TGRLP) and low-density lipoprotein (LDL). ApoM is important for the pre∫-HDL formation. Pre∫-HDL is an important acceptor of peripheral cellular cholesterol. The concentration of ApoM in plasma strongly correlates with total cholesterol. Low concentrations of ApoM in plasma is associated with diabetes.

Description:
Glycosylation of asparagine residues in Asn-X-Ser/Thr motifs in proteins commonly occur in the lumen of the endoplasmic reticulum (ER). Glucosidase I catalyzes the first step in the N-linked oligosaccharide processing pathway. It specifically removes the distal alpha 1,2-linked glucose residue from the Glc3-Man9-GlcNAc2 oligosaccharide precursor. Glucosidase I contains a short cytosolic tail, a single pass transmembrane domain and a large C-terminal catalytic domain located on the luminal side of the ER. Mutations in the gene encoding Glucosidase I result in the congenital disorder glycosylation (CDG-IIb), which is characterized by generalized hypotonia, dysmorphic features, hepatomegaly, hypoventilation, feeding problems, seizures and death. Two point mutations in the Glucosidase I gene have been identified and result in amino acid substitutions, namely Arg486Thr and Phe652Leu, that affect polypeptide folding and active site formation.

Description:
May play a role in tumor progression. Ability to promote tumor growth may be mainly due to repression of apoptosis as opposed to proliferation. Has anti-adhesive properties. Seems to alter cellular behavior through both anti-adhesive effects on cell-cell and cell-extracellular matrix interactions and in its ability to act as an intramembrane ligand for ERBB2. Plays an important role in cell proliferation and differentiation of epithelial cells by inducing specific phosphorylation of ERBB2. The MUC4-ERBB2 complex causes site-specific phosphorylation of the ERBB2 'Tyr-1248'. In polarized epithelilal cells segragates ERBB2 and other ERBB receptors and prevents ERBB2 from acting as a coreceptor. The interaction with ERBB2 leads to enhanced expression of CDKN1B. The formation of a MUC4-ERBB2-ERBB3-NRG1 complex leads to down-regulation of CDKN1B, resulting in repression of apoptosis and stimulation of proliferation.

Description:
LGI4, also known as leucine-rich glioma-inactivated protein 4, is a 537 amino acid secreted glycosylated protein that is widely expressed, with highest levels found within the nervous system. Interestingly, siRNA knockdown studies of LGI4 expression in Schwann cells have been shown to result in the inhibition of myelination, thus suggesting that LGI4 is an essential component of myelin formation and axon segregation. LGI4 shares significant homology with its other family members, LGI1, LGI2 and LGI3. Significantly, mutations in the gene encoding LGI1 have been linked to human temporal lobe epilepsy and, given the sequence similarity of LGI4, it is likely that it also may be implicated in the pathology of seizures. LGI4 is localized subcellularly to the Golgi, ER and vesicles. There are two isoforms of LGI4 that are produced as a result of alternative splicing events.

Description:
RAP80 was initially identified as zinc-finger containing nuclear protein that is highly expressed in testis and interacts with the retinoid-related testis-associated receptor (RTR). Later experiments revealed that RAP80 is recruited by the Coiled-coil domain 98 (CCDC98) protein to the breast cancer-1 protein BRCA1, allowing the formation of BRCA1 foci in response to DNA damage caused by ionizing radiation. Both RAP80 and CCDC98 are required for DNA damage resistance, G2-M checkpoint control, and DNA repair. Cells depleted of either RAP80 or CCDC98 exhibited increased sensitivity to ionizing radiation, although not as much as in BRCA1-depleted cells, suggesting that RAP80 and CCDC98 control only part of the DNA damage response role of BRCA1. At least four isoforms of RAP80 are known to exist.
