Description:
GDNF is a disulfide-linked, homodimeric neurotrophic factor structurally related to Artemin, Neurturin and Persephin. These proteins belong to the cysteine-knot superfamily of growth factors that assume stable dimeric protein structures. GDNF signals through a multicomponent receptor system, composed of a RET and one of the four GFRalpha (alpha1-alpha4) receptors. GDNF specifically promotes dopamine uptake and survival, and morphological differentiation of midbrain neurons. Using a Parkinson's disease mouse model, GDNF has been shown to improve conditions such as bradykinesia, rigidity, and postural instability. The functional rat GDNF ligand is a disulfide-linked homodimer consisting of two 15 kDa polypeptide chains called monomers. Each monomer contains seven conserved cysteine residues, including Cys-101, which is used for inter-chain disulfide bridging, and others that are involved in the intramolecular ring formation known as the cysteine-knot configuration. The calculated molecular weight of Recombinant Rat GDNF is 30.0 kDa.

Description:
Osteoprotegerin (OPG, or osteoclastogenesis inhibitory factor) is a secretory glycoprotein belonging to TNF receptor superfamily. Acts as decoy receptor for RANKL and thereby neutralizes its function in osteoclastogenesis. Inhibits the activation of osteoclasts and promotes osteoclast apoptosis. Bone homeostasis seems to depend on the local RANKL/OPG ratio. May also play a role in preventing arterial calcification. May act as decoy receptor for TRAIL and protect against apoptosis. TRAIL binding blocks the inhibition of osteoclastogenesis. OPG acts as a soluble factor in the regulation of bone mass and may be beneficial in the treatment of osteoporosis with increased osteoclast activity. OPG consists of 401 amino acids with a molecular weight of 44 kDa as a monomer and 90 kDa as a disulphide-linked dimer.

Description:
Zinc-finger proteins contain DNA-binding domains and have a wide variety of functions, most of which encompass some form of transcriptional activation or repression. The majority of zinc-finger proteins contain a Krüppel-type DNA binding domain and a KRAB domain, which is thought to interact with KAP1, thereby recruiting histone modifying proteins. Zinc finger and BTB domain-containing protein 48 (ZBTB48), also known as Kruppel-related zinc finger protein 3 or HKR3, is a 688 amino acid member of the Krüppel C2H2-type zinc-finger protein family. Localized to the nucleus, ZBTB48 is expressed in the adrenal gland and neuroblastoma cell lines. ZBTB48 contains a BTB domain, also known as a POZ domain, which inhibits DNA binding and mediates homotypic and heterotypic dimerization. Characteristics of the BTB domain suggest that ZBTB48 functions as a transcription regulator.

Description:
The peroxisomal membrane contains several ATP-binding cassette (ABC) transporters, ABCD1–4 that are known to be present in the human peroxisome membrane (1). All four proteins are ABC half-transporters, which dimerize to form an active transporter (1). A mutation in the ABCD1 causes X-linked adrenoleukodystrophy (X-ALD), a peroxisomal disorder which affects lipid storage (2,3). ABCD2 in mouse, is expressed at high levels in the brain and adrenal organs, which are adversely affected in X-ALD (4). The peroxisomal membrane comprises 2 quantitatively major proteins, PMP22 and ABCD3 (5). ABCD3 is associated with irregularly shaped vesicles which may be defective peroxisomes or peroxisome precursors (5). ABCD4 localizes to peroxisomes (1). The genes which encode ABCD1–4 map to human chromosome Xq28, 12q11-q12, 1p22-p21 and 14q24.3, respectively (3,6–8). ABCB7 is a half-transporter involved in the transport of heme from the mitochondria to the cytosol and maps to human chromosome Xq13.1-q13.3 (9).

Description:
Cell surface receptor for Reelin (RELN) and apolipoprotein E (apoE)-containing ligands. LRP8 participates in transmitting the extracellular Reelin signal to intracellular signaling processes, by binding to DAB1 on its cytoplasmic tail. Reelin acts via both the VLDL receptor (VLDLR) and LRP8 to regulate DAB1 tyrosine phosphorylation and microtubule function in neurons. LRP8 has higher affinity for Reelin than VLDLR. LRP8 is thus a key component of the Reelin pathway which governs neuronal layering of the forebrain during embryonic brain development. Binds the endoplasmic reticulum resident receptor-associated protein (RAP). Binds dimers of beta 2-glycoprotein I and may be involved in the suppression of platelet aggregation in the vasculature. Highly expressed in the initial segment of the epididymis, where it affects the functional expression of clusterin and phospholipid hydroperoxide glutathione peroxidase (PHGPx), two proteins required for sperm maturation. May also function as an endocytic receptor.

Description:
The telomeric repeat binding factor family (TERF1 and TERF2) plays a key role in cellular immortalization and cellular senescence. TERF2, as a component of the telomere nucleoprotein complex, is present at telomeres in metaphase of the cell cycle. TERF2 is a second negative regulator of telomere length and binds as a dimer to TTAGGG repeats at ends of chromosomes (telomeres), where it blocks inappropriate activation of the ATM/p53 pathway. TERF1 negatively regulates telomere elongation, while TERF2 protects the chromosome ends by inhibiting end-to-end fusions. Down-regulation of TRF expression in tumor cells may contribute to cell immortalization and malignant progression.

Description:
Wnt-induced secreted protein (WISP)-1, WISP-2 and WISP-3 are members of the CCN family of growth factors, which include connective tissue growth factor (CTGF) and Cyr61. WISP-1, WISP-2 and WISP-3 share significant sequence similarity, including four conserved cysteine-rich domains, and they are believed to function as dimers in their active forms. WISP-1 expression is observed in various tissues including adult heart, kidney and spleen, while WISP-2 expression predominates in skeletal muscle, colon and ovary. Both WISP-1 and WISP-2 are upregulated in cells transformed with the proto-oncogene Wnt-1, and they are also more highly expressed in human colon tumors, suggesting that these proteins may participate in tumor development. WISP-3 is involved in normal post-natal skeletal growth, and it is also implicated in the development of the autosomal recessive skeletal disorder progressive pseudorheumatoid dysplasia, which affects cartilage homeostasis by disrupting the growth of chondrocyte and normal cell columnar organization.

Description:
RENBP is a 427 amino acid protein that is a proteinaceous renin inhibitor. In inhibiting renin, RENBP forms a complex with it; a high molecular weight renin. RENBP contains a leucine zipper domain, which is essential for its dimerization with renin. RENBP can catalyze the interconversion of N-acetylglucosamine to N-acetylmannosamine, indicating that it is a GlcNAc 2-epimerase. Sequences of porcine, human and rat renin-binding proteins are highly homologous. The RENBP gene is conserved in dog, mouse, rat and zebrafish, and maps to human chromosome Xq28 between DXS52 and G6PD. Rat Renbp is located on chromosome X at Xq37 close to marker DXWox3 and falls outside the BP QTL regions on chromosome X.

Description:
This gene encodes a member of the bone morphogenetic protein family which is part of the transforming growth factor-beta superfamily. The superfamily includes large families of growth and differentiation factors. Bone morphogenetic proteins were originally identified by an ability of demineralized bone extract to induce endochondral osteogenesis in vivo in an extraskeletal site. These proteins are synthesized as prepropeptides, cleaved, and then processed into dimeric proteins. This protein may act as an important signaling molecule within the trabecular meshwork and optic nerve head, and may play a potential role in glaucoma pathogenesis. This gene is differentially regulated during the formation of various tumors. [provided by RefSeq, Jul 2008].

Description:
Cyclin dependent kinase 5 (Cdk5) is a key regulator of cell cycle progression in neuronal differentiation that physically associates with and is activated by the neuron-specific protein p35. CDK5RAP1 (Cdk5 regulatory subunit-associated protein 1), also known as Cdk5 activator-binding protein C42, is a 601 amino acid protein that specifically inhibits Cdk5 activation by p35 through formation of a dimer that inhibits kinase activity. CDK5RAP1 contains one TRAM domain, which is thought to bind tRNA and deliver the RNA-modifying enzymatic domain to its target. There are 4 named isoforms of CDK5RAP1 that are produced as a result of alternative splicing events and are expressed at high levels in heart and skeletal muscle.
