Xenopus winged-helix factor, xFAST-1 (forkhead activin signal transducer-1) is a transcription factor that forms a complex with the receptor-regulated Smad protein, Smad2, and directly binds to activin response elements on DNA (1,2). The human homolog FAST-1 and the corresponding mouse homolog, designated FAST-2, share significant sequence homology with xFAST-1, including a conserved N-terminal forkhead domain that consists of 110 amino acid residues and is essential for binding DNA and regulating transcription in embryogenesis, in tumorigenesis and in the maintenance of differentiated cell states (3,4). FAST-1 and FAST-2 also contain a distinct C-terminal Smad interaction domain that is required for the association with various Smad proteins, including Smad2, Smad3 and Smad4 (3,5). Expression of FAST-1 and FAST-2 is predominantly observed during early development, with lower levels detected in adult tissues (6,7). FAST-1 and FAST-2 mediated DNA binding is attenuated by both TFGß and activin, indicating that these FAST proteins mediate TFGß induced signal transduction (3).

Tripartite motif-containing protein 34 (TRIM34), also known as RING finger protein 21 (RNF21) or interferon-responsive finger protein 1 (IFP1), is a 488 amino acid member of the TRIM family, also known as the RING-B-box coiled-coil (RBCC) family. Members of the RBCC family have an N-terminal RING finger, followed by one or two zinc-binding domains (B-box domains), a leucine coiled-coil region and a variable C-terminal domain. Three isoforms of TRIM34 exist as a result of alternative splicing events. Isoform 1, the most abundant isoform, is highly expressed in placenta, spleen, colon and peripheral blood leukocytes. Studies have shown that Interferon (IFN) stimulation leads to an upregulation of TRIM34. These findings suggest that TRIM34 maybe a downstream effector that mediates IFN activities.

Nop1p was originally identified as a nucleolar protein of bakers yeast, Saccharomyces cerevisiae. The Nop1p protein is 327 amino acids in size (34.5 kDa), is essential for yeast viability, and is localized in the nucleoli. The systematic name for S. cerevisiae Nop1 is YDL014W, and it is now known to be part of the small subunit processome complex, involved in the processing of pre-18S ribosomal RNA. Nop1p is the yeast homologue of a protein found in all eukaryotes and archea generally called fibrillarin. Fibrillarin/Nop1p is extraordinarily conserved, so that the yeast and human proteins are 67% identical, and the human protein can functionally replace the yeast protein. Patients with the autoimmune disease scleroderma often have strong circulating autoantibodies to a ~34 kDa protein which was subsequently found to be fibrillarin. Recent studies show that knock-out of the fibrillarin gene in mice results in embryonic lethality, although mice with only one functional fibrillarin/Nop1p gene were viable. This antibody is becoming widely used as a convenient marker for nucleoli in a wide variety of species (e.g. 4-6).
