This gene encodes a pleckstrin homology domain-containing adapter protein. The encoded protein is localized to the plasma membrane where it specifically binds phosphatidylinositol 3,4-bisphosphate. This protein may be involved in the formation of signaling complexes in the plasma membrane. Polymorphisms in this gene are associated with age-related macular degeneration. Alternate splicing results in multiple transcript variants. A pseudogene of this gene is found on chromosome 5. (provided by RefSeq, Sep 2010).

Description:
Bub1 was initially identified as the mammalian homolog to the yeast protein through a cDNA screen. Bub1 encodes a kinase involved in the formation of the spindle checkpoint, an important mechanism that ensures high fidelity mitotic chromosome segregation. It is thought to be required for assembly of a functional inner centromere, sister chromatid cohesion via targeting of the Shugoshim protein and metaphase congression. Bub1 functions by phosphorylating cdc20, a member of the mitotic checkpoint complex and activating the spindle checkpoint. A related protein kinase Bub3 interacts with Bub1 and targets it to kinetochores prior to chromosome alignment. Mutations in bub1 have been associated with aneuploidy and several forms of cancer.

Apoptosis, also known as programmed cell death, plays major roles in development and normal tissue turnover in addition to tumor formation. Recently a protein similar to the apoptosis-inducing factor (AIF) was cloned and designated AIFL (also known as AIFM3). AIFM3 is expressed ubiquitously and is predominantly localized to the inner membranes of mitochondria. Unlike AIF, AIFM3 does not translocate to the nucleus upon induction of apoptosis. However, overexpression of AIFM3, like AIF, induced cytochrome c release from the mitochondria, cleavage of caspase 3, and ultimately apoptosis, indicating AIFM3 induces apoptosis through caspase activation. GEFT may play a role in actin cytoskeleton reorganization in different tissues since its activation induces formation of actin stress fibers. GEFT works as a guanine nucleotide exchange factor for the Rho family of small GTPases and links specifically to G alpha q/11-coupled receptors in Rho A activation. GEFT is an important regulator of processes involved in axon and dendrite formation. Involved in skeletal myogenesis, GEFT seems to be an exchange factor primarily for Rac 1 in neurons.
