MYST2 specifically represses AR-mediated transcription. MYST2 is a new AR-interacting protein capable of modulating AR activity. It could play a significant role in regulating AR-dependent genes in normal and prostate cancer cells. The biochemical and genetic interactions of MYST family protein MYST2 with two components of the replication apparatus, MCM2 and ORC1, suggest that MYST2-associated HAT activity may play a direct role in the process of DNA replication. MYST2 is a positive regulatory factor for prereplicative complex assembly.

Description:
TNFAIP3, also known as A20, is located in chromosome band 6q23, a region that is often deleted in B cell lymphomas. Recently, it was identified as a tumor suppressor gene in Hodgkin lymphoma and several subtypes of non-Hodgkin lymphomas. TNFAIP3 was initially identified as a zinc-finger protein that is rapidly and transiently induced by TNF-a, inhibiting NF-kB-dependent gene expression, and protecting cells from TNF-a-cytotoxicity. Overexpression of TNFAIP3 also inhibits the TLR2- and TLR4-mediated interleukin-8 synthesis in airway epithelial cells, suggesting that TNFAIP3 also acts as a negative regulator of TLR-mediated inflammatory responses, thereby protecting the host against harmful over-responses to pathogens. At least two isoforms of TNFAIP3 are known to exist.

Ubiquitin, a highly conserved protein that has a major role in targeting cellular proteins for degradation by the 26S proteosome, is synthesized as a precursor protein consisting of either polyubiquitin chains or a single ubiquitin fused to an unrelated protein. This gene encodes a fusion protein consisting of ubiquitin at the N terminus and ribosomal protein S27a at the C terminus. When expressed in yeast, the protein is post-translationally processed, generating free ubiquitin monomer and ribosomal protein S27a. Ribosomal protein S27a is a component of the 40S subunit of the ribosome and belongs to the S27AE family of ribosomal proteins. It contains C4-type zinc finger domains and is located in the cytoplasm. Pseudogenes derived from this gene are present in the genome. As with ribosomal protein S27a, ribosomal protein L40 is also synthesized as a fusion protein with ubiquitin; similarly, ribosomal protein S30 is synthesized as a fusion protein with the ubiquitin-like protein fubi. LAP3 (leucine aminopeptidase 3), also known as LAPEP or PEPS, is a 519 amino acid protein that localizes to the cytoplasm and belongs to the peptidase M17 family. Existing as a homohexamer, LAP3 uses zinc as a cofactor to catalyze the release of an N-terminal proline from a target peptide and is, therefore, involved in the processing and turnover of intracellular proteins. Multiple isoforms of LAP3 exist due to alternative splicing events. The gene encoding LAP3 maps to human chromosome 4, which houses nearly 6% of the human genome and has the largest gene deserts (regions of the genome with no protein encoding genes) of all of the human chromosomes. Defects in some of the genes located on chromosome 4 are associated with Huntington's disease, Ellis-van Creveld syndrome, methylmalonic acidemia and polycystic kidney disease.

Significantly conserved in chimpanzee, dog, cow, mouse, rat, chicken and zebrafish, PJA2 shares 52% identity with PJA1, which is involved in protein ubiquitination in brain and may play a role in X-linked mental retardation. Encoded by a gene that maps to human chromosome 5q21.3, PJA2 localizes to both endoplasmic reticulum and Golgi apparatus membranes. Participating in E2-dependent, E3 ubiquitin-protein ligase activity, PJA2 binds to a variety of E2s and interacts with ubiquitin-conjugating enzymes, such as UBE2D2, in vitro.
