Description:   Semaphorins are a family of cell surface and secreted proteins that are conserved from insects to humans. Members of this family of proteins are approximately 750 amino acids in length (including signal sequences) and are defined by a conserved extracellular “semaphorin” domain of approximately 500 amino acids containing 14-16 cysteines, blocks of conserved sequences and no obvious repeats. The transmembrane semaphorins are characterized by an additional 80 amino acid transmembrane domain and an 80-110 amino acid cytoplasmic domain. Secreted and cell-bound semaphorins chemically attract and repel the growth of neural axons, guiding the development of intricate networks of neural tissue. SEMA4B (semaphorin-4B), also known as SemC or SEMAC, is an 832 amino acid single-pass type I membrane protein that belongs to the semaphorin family and exists as two alternatively spliced isoforms. Containing one Ig-like C2-type (immunoglobulin-like) domain, a PSI domain and a single sema domain, SEMA4B is encoded by a gene located on human chromosome 15.

Description:   SCAMP4 Antibody: Secretory carrier membrane proteins (SCAMPs) are integral membrane proteins seen in secretory and endocytic carriers that are thought to play roles in membrane trafficking. One member of this family, SCAMP4, lacks the putative calcium binding and leucine zipper structures as well as the protein-protein interacting NPF domains common to most SCAMP family members. In rats, SCAMP4 mRNA is repressed by progesterone in brain regions associated with female sexual behavior. At least three isoforms of SCAMP4 are known to exist. This SCAMP4 antibody is predicted to not cross-react with any other member of the secretory carrier membrane protein family.

Description:   FRMPD2 Antibody: The FERM and PDZ domain containing (FRMPD) protein family consists of four proteins that contain a FERM (Four-point-one, erzin, radixin, moesin) domain and at least one PDZ (PSD-95/Discs large/Zonula-occuldens-1) domain. FRMPD2 also contains an N-terminal KIND domain and three PDZ domains and is structurally similar to the protein tyrosine phosphatase PTP-BL. FRMPD2 is localized in a polarized fashion in epithelial cells at the basolateral membrane and partially co-localizes with the tight-junction marker protein Zonula-occuldens-1. Suppression of FRMPD2 expression via RNAi in Caco-2 cells results in an impairment of tight junction formation, indicating that FRMPD2 plays a major role in tight junction formation. Other experiments indicate that FRMPD2 is a binding partner to several catenin family members and recruitment of FRMPD2 to cell-cell contacts is dependent on E-cadherin-mediated cell-cell adhesion.

Description:   FRMPD1 Antibody: The FERM and PDZ domain containing (FRMPD) protein family consists of four proteins that contain a FERM (Four-point-one, erzin, radixin, moesin) domain and at least one PDZ (PSD-95/Discs large/Zonula-occuldens-1) domain. FRMPD1 was initially identified through a yeast two-hybrid screening with Activator of G-protein signaling 3 (AGS3), a protein that acts to stabilize the GDP-bound conformation of Gai, as the bait. FRMPD1 binds to the tetratricopeptide repeat of AGS3 and stabilizes AGS3 in a membrane environment. Suppression of FRMPD1 in Cath.a-differentiated neuronal cells decreased the level of endogenous AGS3 in membrane fractions by ~50% and enhanced the inhibition of forskolin-induced increases in cAMP. Co-immunoprecipitation studies indicate that the interaction of AGS3 with FRMPD1 is mutually exclusive with the interaction of AGS3 with Gai3, suggesting that FRMPD1 may position AGS3 in the membrane where it can then interact with Galphai.

Description:   TREX2 Antibody: TREX2 is one of two major human 3' to 5' exonucleases which are required for checkpoint signaling after DNA damage. While the related protein TREX1 is required for ATR expression and mutations in this gene result in Aicardi-Goutieres syndrome, chilblain lupus, and Cree encephalitis, less is known about TREX2. Like TREX1, TREX2 is ubiquitously expressed in all tissues examined. X-ray crystallography studies of TREX2-single strand DNA complexes revealed that TREX2 binds DNA as a dimer and may act to displace the second DNA strand, suggesting that TREX2 unzips and denatures double stranded DNA and feed the substrate DNA strand into the TREX2 active site. At least two isoforms of TREX2 are known to exist.

Description:   GPAT1 Antibody: Glycerol-3-phosphate acyltransferase 1 (GPAT1), one of four known GPAT isoforms, is located on the mitochondrial outer membrane, allowing reciprocal regulation with carnitine palmitoyltransferase-1. It is thought to be critical for the development of hepatic steatosis; steatosis triggered by GPAT1 overexpression leads to hepatic and possibly peripheral insulin resistance. GPAT1 is transcriptionally upregulated by insulin and sterol regulatory element binding protein (SREBP-1) and downregulated by AMP-activated protein kinase. Mice deficient in GPAT1 exhibit decreased triacylglycerol (TAG) in cardiomyocytes even in high-fat diets, suggesting that GPAT1 contributes significantly to TAG accumulation in heart tissue during lipogenic or high fat diets.

Description:   PHOX2B Antibody: PHOX2A and PHOX2B (Paired mesoderm homeobox protein) are closely related, paired-homeodomain transcription factors that function as determinants of the noradrenergic phenotype during embryogenesis. PHOX2 proteins are crucial for the regulation of endogenous hydroxylases in neural crest cells and promote sympathetic neuron generation. Human PHOX2B contains one DNA binding homeobox domain and is required for the differentiation of all central and nonperipheral noradrenergic centers in the brain. In contrast, PHOX2A controls only the differentiation of the main noradrenergic center of the brain. Regulation of PHOX2 may have therapeutic utility in aging or disorders involving degeneration of noradrenergic neurons.

Description:   TMEM184C Antibody: Anaplastic thyroid cancer is one of the most lethal forms of cancer, but the precise carcinogenic mechanism has not been identified. TMEM184C, also known as TMEM34, was identified in a cDNA microarray analysis as being down-regulated in anaplastic thyroid cancers compared to normal thyroid tissues. TMEM184C protein expression was also lower in cell lines derived from these types of cancers compared to that of normal thyroid tissues or cell lines based on other types of thyroid cancers. Furthermore, transfection of TMEM34 into KTA2 cells led to the inhibition of cell growth, suggesting that TMEM184C might act as a tumor suppressor in anaplastic thyroid cancers.

Description:   ARHGAP18 Antibody: ARHGAP18 is one member of the human RhoGAP family with approximately 80 RhoGAP proteins known to be encoded in the human genome. Rho proteins belong to the Ras superfamily that is composed of over 50 members divided into six families, including Ras, Sar, Rho, Ran, Rab and Arf. Rho GTPases are important regulators of the actin cytoskeleton and consequently influence the shape and migration of cells. ARHGAP18 is linked to Ras, and thus, to EGFR-mediated proliferation, migration and differentiation. ARHGAP18 is precisely contained within chromosome 6q22-24, which has been shown to be linked to schizophrenia, suggesting that ARHGEP18 may play a role in this condition.

Description:   PTK7 Antibody: Protein-tyrosine kinases (PTKs) play important roles in regulating cell proliferation and differentiation during development. One member of the PTK family, PTK7, has been suggested to regulate the planar cell polarity (PCP) pathway in vertebrates and may play a role in neural convergent extension and neural tube closure. PTK7 has also been implicated in the development of cancer. Loss of PTK7 expression was seen in several melanoma cell lines and biopsies. Conversely, high-throughput analysis of acute myeloid leukemia samples showed an increased level of PTK7 expression compared to normal bone marrow and purified CD34+ cells. Multiple isoforms of PTK7 are known to exist.
