Description:
Catabolite of thymine. Precursor for 3-Hydroxyisobutyric acid synthesis. Small molecule myokine. Browning inducer of white adipose tissue (WAT) into brown adipose tissue (BAT). PGC-1alpha-mediated and exercise-triggered nonadrenergic activator of the thermogenic program in WAT. Increases the expression of brown adipocyte-specific genes (browning) in white adipocytes and beta-oxidation in hepatocytes through a PPARalpha-mediated mechanism. Induces a brown adipose-like phenotype in human pluripotent stem cells and improves glucose homeostasis. Enhances fatty acid oxidation and reduces body weight in mice through an increased production of leptin by the white adipose tissue (WAT). Inhibits Agxt2-mediated metabolism of dimethylarginines. Partial agonist of the glycine receptor (GlyR).

Description:
Methylation at the 5'-position of cytosine is the only known naturally occurring covalent modification of the mammalian genome. DNA methylation requires the enzymatic activity of DNA 5-cytosine methyltransferase (Dnmt) proteins, which catalyze the transfer of a methyl group from S-adenosyl methionine to the 5'-position of cytosines, thereby repressing expression of the target gene. Dnmt3L (DNA (cytosine-5)-methyltransferase 3-like) is a 387 amino acid protein that contains one ADD-type zinc finger and is a member of the Dnmt family. Localized to the nucleus and expressed at lows levels in thymus, testis and ovary, Dnmt3L does not exhibit DNA methyltransferase activity, but is able to stimulate de novo methylation by Dnmt3 and is thought to play a key role in the establishment of genomic imprints. Additionally, Dnmt3L interacts with histone deacetylase 1 (HDAC1) and, through this interaction, mediates transcriptional repression. Multiple isoforms of Dnmt3L exist due to alternative splicing events.

Description:
CpG methylation is an epigenetic modification that is important for embryonic development, imprinting, and X-chromosome inactivation. Studies in mice have demonstrated that DNA methylation is required for mammalian development. DNMT3A is a DNA methyltransferase that is thought to function in de novo methylation, rather than maintenance methylation. The protein localizes to the cytoplasm and nucleus and its expression is developmentally regulated.

Description:
ESET is a nuclear protein belonging to the histone-lysine methyltransferase family and to the Suvar3-9 subfamily. It is a highly conserved protein of 150 amino acids that has been implicated in chromatin structure modulation. ESET is excluded from cell nucleoli and areas of condensed chromatin and can associate with the nonpericentromeric regions of chromatin. The gene encoding for this protein, SETDB1, maps to chromosome 1q21. ESET is a histone methyltransferase, methylating Lys-9 of histone H3 and mutations within the SETDB1 gene abolishes its methyltransferase activity. This methylation acts as a tag for epigenetic transcriptional repression by rounding up HP1 proteins to methylated histones. ESET is widely expressed with highest levels found in testis.

Description:
The synthetic peptide (Tat-Glur23Y) contains tyrosine residues that blocks phosphorylation of alpha-amino-3-hydroxy-5-methyl-isoxazole-4-propionic acid (AMPA) receptor endocytosis. Previous research shows that Tat-Glur23Y blocks regulated AMPA and thereby prevents long-term depression (LTD) in structures such as the nucleus accumbens and dorsal hippocampus.

Description:
Sodium-dependent amino acids transporter that has a broad substrate specificity, with a preference for zwitterionic amino acids. It accepts as substrates all neutral amino acids, including glutamine, asparagine, and branched-chain and aromatic amino acids, and excludes methylated, anionic, and cationic amino acids. May also be activated by insulin. Through binding of the fusogenic protein syncytin-1/ERVW-1 may mediate trophoblasts syncytialization, the spontaneous fusion of their plasma membranes, an essential process in placental development (PubMed:10708449, PubMed:23492904). Acts as a cell surface receptor for feline endogenous virus RD114, baboon M7 endogenous virus and type D simian retroviruses (PubMed:10051606, PubMed:10196349).
