Description:
Betaine is a quaternary amine found in bacteria and higher plants which acts as an osmoprotectant (able to restore and maintain osmotic balance in living cells) and enhances the freezing tolerance of of living organisms. This is probably due to its involvement in the protection of macrocomponents of plant cells, such as protein complexes and membranes, under stress conditions. Betaine is biosynthesized in living cells by a two-step oxidation method from choline. The primary enzyme involved in this biosynthesis is choline oxidase.

Sodium chloride is a commonly used chemical which is found widely in nature. It is considered to be an essential nutrient. Excess amounts of sodium chloride can destroy electrolyte balance and cause death in most animals, including humans. Sodium chloride is used in a wide variety of biochemical applications, including intravenous fluids (normal saline is 0.9% w/v in water), density gradients, a diluent to increase ionic strength in buffers or culture media and in salt-out procedures in the isolation of DNA. It has also been used in high concentrations for preservation of foods since bacteria cannot grow in high salt conditions. Sodium chloride is widely used in protein crystallization.

Phenol red is an acid-base indicator. It is made by condensing two moles of phenol with one mole of o-sulfobenzoic acid anhydride.
Phenol Red is used as a pH indicator in cell culture applications. A solution of phenol red will have a yellow color at a pH of 6.4 or below and a red color at a pH of 8.2 and above. Phenol red in tissue culture media can act as a weak estrogen, especially with human breast cancer cells. Phenol red can be used to measure hydrogen peroxide in cultured macrophages in multiwell plates. Store at Room Temperature (15-30 °C)

Sodium chloride is used in a wide variety of biochemical applications, including intravenous fluids (normal saline is 0.9% w/v in water 10), density gradients 3,6, a diluent to increase ionic strength in buffers or culture media and in salt-out procedures in the isolation of DNA. It has also been used in high concentrations for preservation of foods since bacteria cannot grow in high salt conditions. A salt-ice mixture in the ratio of approximately 33 g sodium chloride to 100 g ice (at -1°C) will drop in temperature to as low as -21°C, depending on the rate of stirring and the size of the ice chunks.

PMA (Phorbol-12-Myristate-13-Acetate) is a diester of phorbol and is a tumor promoting compound extracted from croton oil. It is a reversible, highly potent protein kinase C (PKC) activator in vitro and in vivo at nM concentrations. PMA’s effects on PKC are attributed to its similarity to diacylgylerol, a natural activator of PKC.
PMA activates Ca2+- ATPase and potentiates forskolin-induced cAMP formation. It has been shown to inhibit apoptosis induced by the Fas antigen, but PMA induces apoptosis in HL-60 promyelocytic leukemia cells. Potent tumor promoter; activates protein kinase C in vivo and in vitro. Store at -20 °C. Protect from light.

rProtein A Sepharoseâ„¢ 4 Fast Flow is a well established antibody affinity medium is designed for the purification of monoclonal and polyclonal antibodies based on Sepharoseâ„¢ Fast Flow platform. The recombinant protein A is produced in E.coli and engineered for an oriented coupling to giving a matrix with enhanced binding capacity. The epoxy-based coupling ensures low ligand leakage. The specificity of the recombinant protein A for the Fc region of IgG is similar to native protein A and provides excellent purification in one step. The high capacity, low ligand leakage and a well established base matrix makes rProtein A Sepharoseâ„¢ Fast Flow ideal for purification of monoclonal antibodies at both laboratory and process scale.

Brefeldin A is a fungal metabolite which is a macrocyclic lactone exhibiting a wide range of antibiotic activity. Produced by Penicillium brefeldianum. Blocks binding of the cytosolic coat protein b-COP and ARF to Golgi membranes mediated by protein G. Also blocks protein transportation into post-Golgi compartments. It activates the sphingomyelin cycle. Brefeldin A mediated apoptosis has been observed in human tumor cells. Brefeldin A reversibly inhibits the intracellular translocation of proteins in eukaryotes, e.g., during transport of proteins to the cell surface for secretion or expression. It has been reported to block the response of cultured cells to cholera toxin. In HepG2 cells, BFA induces two blocks in the secretory pathway; one at the level of the endoplasmic reticulum-Golgi juncture and the other in the trans-Golgi network. Brefeldin A is used in the studies of Brefeldin A-inhibited Guanine Nucleotide-exchange Protein, BIG2, Regulates the Constitutive Release of TNFR1 Exosome-like Vesicles. Brefeldin A (BFA) is a fungal metabolite which disrupts the structure and function of the Golgi apparatus. BFA is an activator of the sphingomyelin cycle. Brefeldin A-mediated apoptosis has been observed in human tumor cells.

Glycine is a non-essential amino acid. It is only amino acid with no asymmetric carbon and thus is not chiral. It is the major inhibitory neurotransmitter. It is involved in the biosynthesis of the porphyrin rings of hemes and chlorophylls. Glycine is commonly used in buffer solutions, in electrophoresis and preparative chromatography. A study of the folding of monoclonal antibodies in the presence of glycine and their subsequent purification has been published. The use of glycine in the purification of lipopolysaccharides, lipooligosaccharides, and lipid A has been reported. It is an amino acid for use in cell culture media development applications and existing media formulations. Glycine is commonly used as a component in Tris-glycine and Tris-glycine-SDS running buffers for polyacrylamide gel electrophoresis, a component of Towbin's transfer buffer for Western blots, a buffer substance in cryoenzymology, in osmotic pressure maintenance in isoelectric focusing of erythrocytes, salting-in effect in protein chemistry, and as a buffer component in the coupled phosphatase-kinase reaction for end labelling of restriction fragments. The growth requirements of various microorganisms is reported in the Handbook of Microbiology. Glycine is a non-chiral amino acid that can be synthesized in the body from the amino acid serine by Serine Hydroxymethyltransferase. Inhibitory neurotransmitter in spinal cord, allosteric regulator of NMDA receptors.
