Procaspase-8 belongs to the family of caspases. Binding of FasL to Fas leads to formation of a receptor complex at the cellular membrane, which was named DISC. The DISC consists of oligomerized receptors, the DD-containing adaptor molecule FADD, procaspase-8, procaspase-10 and c-FLIP. The DISC structure provides a platform for the oligomerization of procaspase-8 that allows two procaspase-8 homodimers to be in the close proximity leading to the initial activation of procaspase-8. At the first cleavage step, the N-terminal p43/p41 and the C-terminal p30 cleavage products are generated. Importantly, these cleavage products already possess catalytic activity. At the second cleavage step, p43/p41 and p30 are processed to p10 and p20, respectively, which leads to the generation of the active caspase-8 heterotetramer (p20/p10)2.

FLT3 encodes a class III receptor tyrosine kinase that regulates hematopoiesis. The receptor consists of an extracellular domain composed of five immunoglobulin-like domains, one transmembrane region, and a cytoplasmic kinase domain split into two parts by a kinase-insert domain. The receptor is activated by binding of the fms-related tyrosine kinase 3 ligand to the extracellular domain, which induces homodimer formation in the plasma membrane leading to autophosphorylation of the receptor. The activated receptor kinase subsequently phosphorylates and activates multiple cytoplasmic effector molecules in pathways involved in apoptosis, proliferation, and differentiation of hematopoietic cells in bone marrow. Mutations that result in the constitutive activation of this receptor result in acute myeloid leukemia and acute lymphoblastic leukemia.

Syntaphilin Antibody: Syntaphilin was initially identified in a yeast two-hybrid screen with the carboxy terminal region of Syntaxin-1 as bait. Syntaxin-1 is a key component of the synaptic vesicle docking machinery that forms the SNARE complex with synaptobrevin and SNAP-25. Syntaphilin competes with SNAP-25 for binding to syntaxin-1 and inhibits the formation of the SNARE complex, thereby potentially regulating synaptic vesicle exocytosis. Syntaphilin also binds dynamin-1 and inhibits dynamin-dependent endocytosis. Mice lacking syntaphilin show an increased level of mitochondrial motility and a reduced density of axonal mitochondria. This correlates with an enhanced short-term facilitation and significant impairments in motor ability, suggesting syntaphilin plays a major role in presynaptic function. Multiple isoforms are known to exist.

SIP1 Antibody: SIP1 is one of the proteins found in the SMN complex, which consists of the survival of motor neuron (SMN) protein and several gemin proteins. The SMN complex is localized to a subnuclear compartment called gems (gemini of coiled bodies) and is required for assembly of spliceosomal snRNPs and for pre-mRNA splicing. SIP1 interacts directly with the SMN and it is required for formation of the SMN complex. A knockout mouse targeting the mouse homolog of this gene exhibited disrupted snRNP assembly and motor neuron degeneration. However, knockdown of the SIP1 mRNA in motor neurons showed normal motor axons while that of SMN mRNA did show abnormal motor axon outgrowth, indicating that SIP1 may have additional roles outside of the SMN complex.
