Albumin is a soluble, monomeric protein which comprises about one-half of the blood serum protein. Albumin functions primarily as a carrier protein for steroids, fatty acids, and thyroid hormones and plays a role in stabilizing extracellular fluid volume. Mutations in this gene on chromosome 4 result in various anomalous proteins. Albumin is a globular unglycosylated serum protein of molecular weight 65000. The human albumin gene is 16961 nucleotides long from the putative 'cap' site to the first poly(A) addition site. It is split into 15 exons which are symmetrically placed within the 3 domains that are thought to have arisen by triplication of a single primordial domain. Albumin is synthesized in the liver as preproalbumin which has an N-terminal peptide that is removed before the nascent protein is released from the rough endoplasmic reticulum. The product, proalbumin, is in turn cleaved in the Golgi vesicles to produce the secreted albumin.

β-NADP is a coenzyme necessary for the alcoholic fermentation of glucose and the oxidative dehydrogenation of other substances. It occurs widely in living tissue, especially in the liver. Nicotinic acid can be converted to nicotinamide in the body and, in this form, is found as a component of two oxidation-reduction coenzymes: nicotinamide adenine dinucleotide (NAD) and nicotinamide adenine dinucleotide phosphate (NADP). The nicotinamide portion of the coenzyme transfers hydrogens by alternating between oxidized quaternary nitrogen and a reduced tertiary nitrogen. The Hoechst stains are a family of fluorescent stains for labeling DNA in fluorescence microscopy. The blue fluorescent Hoechst dye is a cell permeable nucleic acid stain that has multiple applications, including sensitive detection of DNA in the presence of RNA in agarose gels, automated DNA determination, sensitive determination of cell number and chromosome sorting. Useful vital stain for the flow cytometric recognition of DNA damage and other viability measurements by monitoring the emission spectral shifts of the dyes. Because this fluorescent stain labels DNA, it can also be used to visualize nuclei and mitochondria. Hoechst 34580 can be excited by violet laser at 405 nm, and emit blue/cyan fluorescence light around an emission maximum at ~440 nm. Cyclooxygenases-1 and 2 (COX-1 and COX-2), also known as prostaglandin endoperoxide H synthases-1 and 2, are integral membrane proteins which catalyze the first step of prostaglandin, thromboxane and prostacyclin production from arachidonic acid. Both COX-1 and COX-2 catalyze a cyclooxygenase (bis-oxygenase) reaction in which arachidonate plus two molecules of O2 are converted to PGG2 (prostaglandin G2), and a peroxidase reaction in which PGG2 undergoes a two-electron reduction to PGH2. COX-1 and COX-2 proteins are encoded by separate genes located on different chromosomes, and both enzymes are very similar in structure. They are homodimeric, heme containing, glycosylated proteins with two catalytic sites, but they differ substantially in their pattern of expression and biology. COX-1 is expressed constitutively in most tissues and at high levels in selected cells and tissues, including endothelium, monocytes, platelets, renal collecting tubules and seminal vesicles. COX-1 is thought to regulate a number of housekeeping activities involved in renal, gastrointestinal and platelet function. Inducible COX-2 is undetectable in most mammalian tissues but its expression can be induced rapidly (2-6h) in fibroblasts, endothelial cells, monocytes and ovarian follicles in response to growth factors, tumor promoters, hormones, bacterial endotoxin and cytokines. COX-2 is also expressed constitutively in specialized cell types or tissues where it plays specific functions in individual biological processes. These include reproduction, immunity, renal physiology, neurotransmission, bone resorption and pancreatic secretion. The COX-1 and COX-2 enzymes are important pharmacologically as targets of nonsteroidal anti-inflammatory drugs (NSAIDs) including aspirin, ibuprofen and the COX-2 inhibitors. Inhibition of cyclooxygenases with NSAIDs acutely reduces inflammation, pain and fever. Some methods to determine COX activity include measuring uptake of oxygen using an oxygraph, measuring the conversion of radioactive arachidonic acid, or measuring prostaglandins formed from PGH2 (such as determining PGH2 using immunoassays). These methods can be complex, time consuming and prone to interferences.
