Description:
The solute carrier family SLC35 consists of at least 17 proteins that act as nucleotide sugar transporters localized to the Golgi apparatus and endoplasmic reticulum. The novel protein SLC35D3 is highly homologous to SLC35D1 and SLC35D2, both of which transport UDP-glucuronic acid and UDP-N-acetylgalactosamine, suggesting that SLC35D3 is also involved in the transport of nucleotide sugars. It has been suggested that SLC35D3 regulates platelet dense granules, lysosome-related organelles which contain high concentrations of several biologically important low molecular weight molecules necessary for normal blood homeostasis.

EBI3 is a subunit in two distinct heterodimeric cytokines: interleukin-27 (IL-27) and IL-35. Like interleukin-23 (IL-23), IL-27 is a recently discovered member of the IL-6/IL-12 family of proinflammatory and immunoregulatory cytokines. It exists as a heterodimer composed of the p40-related protein EBI3 and an IL-12 p35-related protein termed p28. IL-27 is produced after activation by antigen-presenting cells and induces proliferation of naïve but not memory CD4+ T-cells. It acts by binding to its receptor WSX-1 (also known as TCCR) and gp130 which results in the activation of a Jak/STAT signaling cascade, suggesting the IL-27 is involved in the regulation of immune processes. It has been suggested that IL-27 can also be used as a therapeutic agent against cancer as it can also induce tumor-specific anti-tumor activity mediated through CD8+ T-cells, IFN-gamma, and T-bet. IL-35 is composed of EBI3 and the p35 subunit of IL-12 and has been reported to have therapeutic effects against collagen-induced arthritis by expanding the population of regulatory T cells and suppressing Th17 cells. At least two isoform of EBI3 are known to exist.

A sensitive reagent for measuring the free sulfhydryl content in proteins, peptides, and tissues. Used to characterize reactive thiol groups and photometric determination of thiols and for measuring low-molecular mass thiols such as glutathione in both pure solutions and biological samples, such as blood. It can also measure the number of thiol groups on proteins. Through reaction with aliphatic thiol groups a mixed disulfide of protein thiol and one mole of 2-nitro-5-thiobenzoate per mole of protein sulfhydryl group is being formed. DTNB has little absorbance. Reaction with -SH groups on proteins (from any solvent accessible Cys) under mild alkaline conditions (pH 7-8) produces the 2-nitro-5-thiobenzoate anion, which gives an intense yellow color with an absorption maximum at 409.5nm (Extinction coefficient: 14150 M-1*cm-1). Sensitive to various buffer ions, therefore, the extinction coefficient used to calculate the number of sulfhydryl groups must be matched to the reaction conditions. In case the thiol groups are in disulfide bonds, they must be reduced under anaerobic conditions prior to reaction with DTNB.

IL12B is a subunit of interleukin 12, a cytokine that acts on T and natural killer cells, and has a broad array of biological activities. Interleukin 12 is a disulfide-linked heterodimer composed of the 40 kD cytokine receptor like subunit encoded by this gene, and a 35 kD subunit encoded by IL12A. This cytokine is expressed by activated macrophages that serve as an essential inducer of Th1 cells development. This cytokine has been found to be important for sustaining a sufficient number of memory/effector Th1 cells to mediate long-term protection to an intracellular pathogen. Lectins, of either plant or animal origin, are carbohydrate binding proteins that interact with glycoprotein and glycolipids on the surface of animal cells. The Galectins are lectins that recognize and interact with beta-galactoside moieties. Galectin-3 regulates a number of biological processes, including embryogenesis, inflammatory responses, cell progression and metastasis. Galectin-3 is normally expressed in epithelia of a variety of tissues, including colon and endometrium, and in various inflammatory cells, including macrophages. Galectin-3 can function intracellularly, in controlling cell cycle and preventing T-cell apoptosis, and also extracellularly, in activating various cells, including monocytes/macrophages, mast cells, neutrophils, and lymphocytes. Expression of Galectin-3 is affected by neoplastic transformation, being up-regulated in certain types of lymphomas, and in thyroid and hepatic carcinomas. Conversely, it is down-regulated in other cancers, such as colon, breast, ovarian, and uterine. Recombinant human Galectin-3 is a globular 26.0kDa protein containing 250 amino acid residues, but no disulfide bonds.

The Hoechst stains are a family of fluorescent stains for labeling DNA in fluorescence microscopy. The blue fluorescent Hoechst dye is a cell permeable nucleic acid stain that has multiple applications, including sensitive detection of DNA in the presence of RNA in agarose gels, automated DNA determination, sensitive determination of cell number and chromosome sorting. Useful vital stain for the flow cytometric recognition of DNA damage and other viability measurements by monitoring the emission spectral shifts of the dyes. Because this fluorescent stain labels DNA, it can also be used to visualize nuclei and mitochondria. Hoechst 34580 can be excited by violet laser at 405 nm, and emit blue/cyan fluorescence light around an emission maximum at ~440 nm. It is cell-permeable and an excellent tool for flow cytometry studies.
