B lymphocytes marker CD20 is a non glycosylated protein with a molecular weight of 35 or 37 kDa depending on the degree of phosphorylation. Although not a member of the tetraspanin superfamily of cell surface receptors, it crosses the cell membrane four times. The CD20 antigen is present on human pre B lymphocytes and on B lymphocytes at all stages of maturation, except on plasma cells. Low level expression of the CD20 antigen has been detected on normal T lymphocytes. This antibody recognizes a protein of 18-35 kDa, identified as CD90 (also known as Thy1). CD90 is a member of the immunoglobulin superfamily. It may contribute to inhibition of proliferation/differentiation of hematopoietic stem cells and neuron memory formation in the CNS. It consists of a single Ig domain (112 amino acids; 25-35 kDa) inserted into the cell membrane via a GPI anchor. Expressed by hematopoietic stem cells and neurons in all species studied. Its highly expressed in connective tissue and various fibroblast and stromal cell lines, expressed on all thymocytes and peripheral T cells in mice, but in humans expressed only on small % fetal thymocytes, 10-40% of CD34 cells in bone marrow, and <1% of CD3 CD4 lymphocytes in peripheral circulation. It is also expressed by human lymph node HEV endothelium but not other endothelia. Lastly, it is expressed by a limited number of lymphoblastoid and leukemic cell lines.

This antibody reacts with adenovirus. The adenovirus early gene product E1A is a potent stimulator of cellular proliferation, which when overexpressed can overcome the growth-inhibitory effects of TGF-β. The E1A region encodes a series of related proteins (35-46 kDa) with multifunctional capabilities and forms a specific complex with the retinoblastoma tumor suppressor gene product. The E1A and E1B regions together comprise the transforming region of adenovirus. While expression of E1A alone is sufficient to immortalize primary cells, complete transformation requires the additional expression of the E1B region.

The protein is important for maintaining normal membrane structure and supporting cell surface protein function. Alpha Fodrin is one of the important targets cleaved by caspases during apoptosis. The full length 240 kDa protein can be cleaved at several sites within its sequence by activated caspases generating N terminal 150 kDa, C terminal 120 kDa, and 35 kDa major products.
