Tyrosinase is the key enzyme for melanin,synthesis in mammalian melanocytes and has been considered to be a unique marker for the study of melanocyte differentiation. A cDNA library was constructed from poly(A)+ mRNA from mouse melanocytes and screened using anti- tyrosinase antiserum and oligonucleotide probes corresponding to amino acid sequence of tyrosinase. sequencing of some cDNA clones positive in these screenings gave a nucleotide sequence of 1838 nucleotides including a open reading frame of 1344 nucleotides that was found to correspond exactly to the amino acid sequence of the cyanogen bromide fragments of tyrosinase.

The forkhead domain-containing gene family (Fox) comprises over 20 members in mammals and is defined by a conserved 110 amino-acid motif containing a winged helix structure DNA-binding domain. The members of this gene family have been implicated as key regulators of embryogenesis, cell cycling, cell lineage restriction and cancer. As such, FOXN2 contains a domain with homology to the forkhead DNA binding domain. FOXN2, or Human T-cell leukemia virus enhancer factor, is a 341 amino acid protein mapping to human gene FOXN2, which has been localized to human chromosome 2p16-p22. This protein, encoded by a 1239-bp cDNA isolated from the Jurkat cDNA library, is capable of binding to a region of the human T-cell leukemia virus long terminal repeat (HTLV-I LTR) located between amino acids 155 and 117. This purine-rich region is important in the regulation of gene expression by the Ets family of transcription factors. FOXN2 is a unique cellular gene that may function in the transcriptional regulation of HTLV-I LTR.

Myostatin is a TGF-β family member that acts as an inhibitor of skeletal muscle growth. This muscle-specific cytokine interacts with Activin type I and type II receptors, and suppresses myoblast proliferation by arresting cell-cycle in the G1 phase. Suppression of myostatin activity facilitates muscle formation, and may be useful in reducing and/or preventing adiposity and type-2 diabetes. Myostatin activity can be blocked by the activin-binding protein follistatin, and by the propeptide of myostatin. The amino acid sequence of mature myostatin is extremely conserved across species, and is the same in murine, rat, chicken, turkey, porcine, and human. Myostatin is expressed as the C-terminal part of a precursor polypeptide, which also contains a short N-terminal signal sequence for secretion, and a propeptide of 243 amino acids. After dimerization of this precursor, the covalent bonds between the propeptide and the mature ligand are cleaved by furin-type proteases. However, the resulting two proteins remain associated through non-covalent interactions, and are secreted as a latent complex. Recombinant Human/Murine/Rat Myostatin is a 25.0 kDa protein consisting of two identical 109 amino acid polypeptides linked by a single disulfide bond.

ILVBL is a 632 amino acid single-pass membrane protein that belongs to the TPP enzyme family. Expressed in the majority of tissues, ILVBL has the highest level of expression in heart, pancreas and placenta. ILVBL is highly homologous to several bacterial enzymes, including the B isozyme of the large catalytic subunit of E. coli acetohydroxy-acid synthase (AHAS) and the oxalyl-coA decarboxylase of O. formigenes, that utilize thiamine pyrophosphate as a cofactor. ILVBL binds one magnesium ion and one thiamine pyrophosphate per subunit, and may catalyze the initial step in branched-chain amino acid biosynthesis. The gene encoding ILVBL maps to human chromosome 19p13.12 and mouse chromosome 10 C1.

BOLA1 (BolA-like protein 1), also known as CGI-143, is a member of the BolA/yrbA family of proteins. Members of this family are homologs of the Escherichia coli protein, BolA. BolA-like proteins are evolutionarily conserved from prokaryotes to eukaryotes and are believed to play a role in cell-cycle regulation or cell proliferation possibly via some sort of transcription regulation of other genes. In addition, BolA-like proteins may contain nucleic-acid binding properties, as is suggested by a fold structure that is similar to the KH-fold, a motif known to participate in nucleic-acid binding. Characteristic of BolA-like proteins which typically consist of approximately 100 amino acids, BOLA1 is a 137 amino acid protein.
