Description:
The NOK-1 monoclonal antibody specifically reacts with human CD178, which is the CD95 or Fas ligand. CD178 is a TNF superfamily type II transmembrane glycoprotein expressed by activated T and NK cells and is involved in Fas-mediated apoptosis of lymphocytes. CD178 is also expressed by monocytes, neutrophils, granulocytes and the parenchymal cells of the retina and cornea. The NOK-1 antibody has been reported to bind to COOH-terminus of the Fas ligand in the region associated with Fas binding.

The mediator complex is a multi-protein transcriptional co-activator that is expressed ubiquitously in eukaryotes from yeast to mammals and is required for induction of RNA polymerase II (pol II) transcription by DNA binding transcription factor. One of the proteins in this complex is MED28, also known as Magicin. MED28 is expressed in many cell lines and tissues. It has been shown that a down-regulation of MED28 expression in NIH3T3 cells results in a significant induction of several genes associated with smooth muscle cell differentiation and conversely its over-expression represses expression of SMC genes. MED28 can also form a ternary complex with Grb2 and Merlin, the neurofibromatosis 2 tumor suppressor protein, indicating that MED28 may play a role in Merlin's tumor suppressive activity. MED28 has also been recently identified as an HIV dependency factor (HDF), suggesting that MED28 may be an important drug target in HIV treatment. At least two isoforms of MED28 are known to exist.

Keratins are a family of intermediate filament proteins that assemble into filaments through forming heterodimers of one type I keratin (keratins 9 to 23) and one type II keratin (keratins 1 to 8). Keratins demonstrate tissue and differentiation specific expression profiles. Keratin 15 is a type I keratin which is expressed only in basal keratinocytes in stratified epithelia and does not appear to have a natural type II expression partner. Keratin 15 is down regulated in activated keratinocytes. Cytokeratin 15 is a specific marker of stem cells of the hair-follicle bulge and may be a useful marker for diagnosis between basal cell carcinoma (BCC) and trichoepithelioma. Trichoblastoma are benign neoplasms of follicular differentiation frequently found in nevus sebaceous. Many morphologic features are shared with nodular basal cell carcinoma, sometimes rendering a diagnosis difficult. Trichoblastoma and BCC show variable expression of Cytokeratin 15 and Cytokeratin 19, and absence of hair keratins.

There are two forms of glutamic acid decarboxylases (GADs) that exist in brain: GAD-65 (also known as GAD2) and GAD-67 (also known as GAD1, GAD or SCP). GAD-65 and GAD-67 are members of the group II decarboxylase family of proteins and are responsible for catalyzing the rate limiting step in the production of GABA (gamma-aminobutyric acid) from L-glutamic acid. Although both GADs are found in brain, GAD-65 localizes to synaptic vesicle membranes in nerve terminals, while GAD-67 is distributed throughout the cell. GAD-67 is responsible for the basal levels of GABA synthesis. In the case of a heightened demand for GABA in neurotransmission, GAD-65 transiently activates to assist in GABA production.
