The 3-HAO family of proteins consists of intramolecular dioxygenases that contain non-heme ferrous iron. HAAO (3-hydroxyanthranilate 3,4-dioxygenase), also known as HAO or 3-HAO, is a 286 amino acid monomeric cytoplasmic protein that belongs to the 3-HAO family. Utilizing Fe2+ ions as a cofactor, HAAO participates in the formation of quinolinic acid (QUIN), and excitotoxin, by catalyzing the oxidative ring opening of 3-hydroxyanthranilate to 2-amino-3-carboxymuconate semialdehyde. HAAO may play a role in disorders that are associated with varying levels of QUIN expression, and is widely expressed in peripheral organs, including kidney and liver. Existing as two alternatively spliced isoforms, the gene encoding HAAO maps to human chromosome 2p21 and mouse chromosome 17 E4.

While PTH acts to increase the concentration of ionic calcium (Ca2+) in the blood, calcitonin, a hormone produced by the parafollicular cells (C cells) of the thyroid gland, acts to decrease ionic calcium concentration. PTH essentially acts to increase the concentration of calcium in the blood by acting upon the parathyroid hormone 1 receptor, which is present at high levels in bone and kidney, and the parathyroid hormone 2 receptor, which is present at high levels in the central nervous system, pancreas, testis, and placenta. The human ataxin-7 gene, also known as spinocerebellar ataxia 7 or SCA7, maps to chromosome 3p13-p12, has a 2,727-bp open reading frame, and encodes a 892 amino acid protein containing a nuclear localization signal and a polyglutamine tract (1,2). SCA7 is an autosomal dominant neurodegenerative disorder characterized by ataxia and selective neuronal cell loss caused by the expansion of a translated CAG repeat encoding a polyglutamine tract in ataxin-7, which is the SCA7 gene product (3,4). Ataxin-7 is expressed within neurons both affected and unaffected in SCA7 pathology with subcellular localization being variable depending upon the neuronal subtype (5). Polyglutamine expanded in ataxin-7 may carry out its pathogenic effects in the nucleus by altering the matrix-associated nuclear structure and/or by disrupting nucleolar function (6).
