HSP 40-4, also known as HDJ2, is the human DnaJ homolog that functions as a co-chaperone with a cysteine-rich zinc finger domain. The cellular redox enzyme thioredoxin interacts with HSP 40-4, and oxidation and reduction reversibly regulate HSP 40-4 function in response to the changing redox states of the cell. The zinc finger domain of HSP 40-4 may act as a redox sensor of chaperone-mediated protein-folding machinery, since HSP 40-4 inactivation leads to the oxidation of cysteine thiols and a simultaneous release of coordinated zinc. Loss of the HSP 40-4 protein may be linked to severe defects in spermatogenesis that involve aberrant androgen signaling.

Aldh3A2 is a member of the aldehyde dehydrogenase superfamily, a group of NAD(P)(+)-dependent enzymes that catalyze the oxidation of a wide spectrum of aliphatic and aromatic aldehydes. Aldehyde dehydrogenase enzymes are thought to play a major role in the detoxification of aldehydes generated by alcohol metabolism and lipid peroxidation. Aldh3A2 catalyzes the oxidation of long-chain aliphatic aldehydes to fatty acids. Mutations in the Aldh3A2 gene cause Sjogren-Larrson syndrome, an inherited neurocutaneous disorder. Patients with this disorder display ichthyosis, mental retardation and spastic diplegia. The pathogenesis of the cutaneous and neurological symptoms is thought to result from abnormal lipid accumulation in the membranes of skin and brain, the formation of aldehyde Schiff base adducts with amine-containing lipids or proteins, or defective eicosanoid metabolism.

Ferritin is a large, iron-storage heteropolymeric protein,which is expressed in most kinds of cells and co-assemble in different proportion in a tissue-specific manner. Ferritin has oligomer of 24 subunits and two types of subunits including light chain(FTL) and heavy chain. Ferritin can remove Fe (â…¡) from solution in the presence of oxygen and is very important for iron homeostasis. Iron is absorbed in the ferrous form and deposited as ferric hydroxides after oxidation. Iron is first oxidized to the ferric state for storage as ferric oxyhdroxide whithin the protein shell of ferritin. Thus, ferritin removes excess iron from the cell sap where it could otherwise participate in peroxidation mechanisms. Ferritin also plays a role in delivery of iron to cells and mediates iron uptake in capsule cells of the developing kidney.
