Description:
Insulin-like growth factor 1 (IGF-1) is also known as somatomedin C, IGF1A, IGFI, sulfation factor, and is a hormone similar in molecular structure to insulin. It plays an important role in childhood growth and continues to have anabolic effects in adults. A synthetic analog of IGF-1, mecasermin is used for the treatment of growth failure. IGF-1 consists of 70 amino acids in a single chain with three intramolecular disulfide bridges. IGF-1 has a molecular weight of 7649 daltons. IGF-1 is produced primarily by the liver as an endocrine hormone as well as in target tissues in a paracrine/autocrine fashion. IGF-1 binds to at least two cell surface receptors: the Insulin-like growth factor 1 receptor, abbreviated as "IGF1R", and the insulin receptor. The IGF-1 receptor seems to be the "physiologic" receptor - it binds IGF-1 at significantly higher affinity than the IGF-1 that is bound to the insulin receptor. Like the insulin receptor, the IGF-1 receptor is a receptor tyrosine kinase - meaning it signals by causing the addition of a phosphate molecule on particular tyrosines. Its primary action is mediated by binding to its specific receptor IGF1R, present on many cell types in many tissues. Binding to the IGF1R, a receptor tyrosine kinase, initiates intracellular signaling; IGF-1 is one of the most potent natural activators of the AKT signaling pathway, a stimulator of cell growth and proliferation, and a potent inhibitor of programmed cell death. Insulin-like growth factor 1 has been shown to bind and interact with all the IGF-1 Binding Proteins (IGFBPs), of which there are six (IGFBP1-6). Specific references are provided for interactions with IGFBP3, IGFBP4 and IGFBP7.

Description:
Involved in several stages of intracellular trafficking. Interacts with membranes containing phosphatidylinositol 3-phosphate (PtdIns(3P)) or phosphatidylinositol 3,5-bisphosphate (PtdIns(3,5)P2) (PubMed:12198132). Acts in part as component of the retromer membrane-deforming SNX-BAR subcomplex. The SNX-BAR retromer mediates retrograde transport of cargo proteins from endosomes to the trans-Golgi network (TGN) and is involved in endosome-to-plasma membrane transport for cargo protein recycling. The SNX-BAR subcomplex functions to deform the donor membrane into a tubular profile called endosome-to-TGN transport carrier (ETC) (Probable). Can sense membrane curvature and has in vitro vesicle-to-membrane remodeling activity (PubMed:19816406, PubMed:23085988). Involved in retrograde endosome-to-TGN transport of lysosomal enzyme receptors (IGF2R, M6PR and SORT1) and Shiginella dysenteria toxin stxB. Plays a role in targeting ligand-activated EGFR to the lysosomes for degradation after endocytosis from the cell surface and release from the Golgi (PubMed:12198132, PubMed:15498486, PubMed:17550970, PubMed:17101778, PubMed:18088323, PubMed:21040701). Involvement in retromer-independent endocytic trafficking of P2RY1 and lysosomal degradation of protease-activated receptor-1/F2R (PubMed:16407403, PubMed:20070609). Promotes KALRN- and RHOG-dependent but retromer-independent membrane remodeling such as lamellipodium formation; the function is dependent on GEF activity of KALRN (PubMed:20604901). Required for endocytosis of DRD5 upon agonist stimulation but not for basal receptor trafficking (PubMed:23152498).

Description:
This MAb recognizes the double stranded DNA in human cells. It can be used to stain the nuclei in cell or tissue preparations and can be used as a nuclear marker in human cells. This MAb produces a homogeneous staining pattern in the nucleus of normal and malignant cells. Deoxyribonucleic acid (DNA) is a nucleic acid that stores long-term information regarding the development and function of all known living organisms. DNA consists of two long nucleotide polymers, which are composed of four bases, namely adenine, thymine, guanine and cytosine, all of which are flanked by a phosphate-deoxyribose backbone. Normally, DNA exists as a double-stranded (ds) molecule that forms in the shape of a double helix, allowing the bases and the backbone of the two strands to interact, thus forming a polynucleotide. When the double helix is unwound (either by enzymes or heat), DNA exists as a single-stranded (ss) molecule that is less stable than the double helix, but is necessary for protein access to DNA bases. Double stranded DNA markers are useful tools in biology research and aid in the study of DNA behavior and characteristics.
CF® dyes are Biotium's next-generation fluorescent dyes. CF®488A is a green fluorescent dye (Ex/Em 490/515 nm) with excellent brightness and photostability. The dye is minimally charged for less non-specific binding. CF®488A also is compatible with super-resolution imaging by TIRF.

Description:
The octapeptide circulatory hormone Angiotensin II is a critical regulator of cardiovascular homeostasis formed in blood, with dual function in tissues of the brain, kidney, heart, and vasculature as an autocrine/paracrine growth hormone. The actions of angiotensin II are mediated by type one (AT1) and type two (AT2) angiotensin II receptors, members of the seven-transmembrane Gprotein coupled receptor (GPCR) family. AT1 receptors couple predominantly with Gq/11 alpha subunits, and signal through phospholipases C, D, and A2, inositol phosphates, and calcium channels, as well as a variety of serine/threonine and tyrosine kinases. AT1 expression is greatest in vascular smooth muscle, liver, kidney, adrenal cortex, brain, and lung, and is primarily responsible for the physiological effects of angiotensin II, including regulation of arterial blood pressure, electrolyte and water balance, thirst, hormone secretion, and renal function. The AT2 receptor displays approximately 30% identity to the AT1 receptor, and is highly expressed during embryonic development. AT2 expression in adult tissue is limited to the adrenal medulla, brain, and reproductive tissues, but is upregulated in injured tissue following myocardial infarction, vascular injury, and wound healing. AT2 receptor signaling is not dependent on G-protein coupling or receptor phosphorylation, although coupling to Gia2 and Gia3 alpha subunits occurs in some tissues. AT2 activation can stimulate tyrosine and serine/threonine phosphatases through activation of nitric oxide and cGMP, and appears to counteract the stimulatory effects of AT1 receptor and growth factor receptor signaling.

The J116 monoclonal antibody specifically reacts with human Programmed death-1 (PD-1 or CD279), a 50-55 kDA glycoprotein. It is expressed on mainly on activated B, T, and myeloid cells. Within the cytoplasmic region, PD-1 contains an Immunoreceptor tyrosine-based inhibitory motif (ITIM) and seems to regulate peripheral tolerance. The lack or mutation of CD279 is linked to autoimmune disorders. The J116 antibody does not block the binding of the PD-1 ligand, but does inhibit the PD-1 signal transduction.

The 17A2 monoclonal antibody specifically reacts with the mouse T lymphocytes receptor (TCR) associated CD3 complex, resulting in cellular activation and proliferation. CD3 is expressed by thymocytes and mature lymphocytes, and contains γ, δ, and ε subunits, involved in the assembly, trafficking, and surface expression of T-cell receptor complex. The interaction between the T lymphocytes and the 17A2 antibody can be blocked by the 145-2C11 anti-CD3e antibody, demonstrating that the 17A2 antibody recognizes the CD3 ε chain.

The C17.8 antibody specifically reacts with the p40 subunit of IL-12, as free monomer, as a homodimer, or as a part of the p70 dimer. C17.8 seems to neutralize the activity of mouse IL-12 and IL-23. IL-12 is the p70 heterodimer of p35 and p40, expressed by the macrophages, dendritic cells, and monocytes. The C17.8 antibody cross-reacts with IL-23, since IL-23 also contains one p40 subunit.
