Bentamapimod (AS602801) is a novel and orally active c-Jun-N-Terminal Kinase Inhibitor highly specific for JNK kinases. The inhibitor Bentamapimod (AS602801) is a potent anti-inflammatory agent which has been shown to reduce the disease burden in several pre-clinical models for endometriosis, and has the potential to be effective in addressing the causes of post-operative abdominal adhesions. Pre-clinical data have demonstrated Bentamapimod to be an effective anti-inflammatory compound with anti-fibrotic properties. Bentamapimod (AS602801) showed good bioavailability in rats (F=50%). It does cross the blood brain barrier, with 98% brain penetration the drug has been well tolerated so far. Bentamapimod (AS602801) is a potent and selective JNK inhibitor with therapeutic potential in MS and fibrosis as well.

Quizartinib (AC220) is a second-generation FLT3 inhibitor that has shown promising activity in AML in Phase II clinical trials. It inhibits mutant and wild-type FLT3 in vivo at 0.1 and 0.5µM, respectively, and has shown favorable activity and tolerability in phase I and II trials in acute myeloid leukemia, with QT prolongation as the dose-limiting toxicity. Recently, AC220 has also been shown to be an effective inhibitor of ATP-binding cassette (ABC) proteins ABCB1 (P-glycoprotein) and ABCG2 (breast cancer resistance protein). Quizartinib inhibited transport of fluorescent ABCG2 and ABCB1 substrates in ABCG2- and ABCB1-overexpressing cells in a concentration-dependent manner, from 0.1 to 5µM and from 0.5 to 10µM, respectively, and inhibited-IAAP photolabeling of ABCG2 and ABCB1 with IC(50) values of 0.07 and 3.3µM, respectively.

The BCR-ABL oncogene triggers intracellular signaling, activating multiple transduction cascades, promoting the growth, proliferation and survival of hematopoietic cells. BCR-ABL plays a role in defective DNA repair, alteration of cellular adhesion and inhibition of apoptosis. Degregulated BCR-ABL tyrosine kinase activity is the molecular marker for CML. Inhibiting BCR-ABL tyrosine kinase activity leads to the induction of apoptosis and inhibits cellular proliferation in vitro and there are various BCR-ABL isoforms with differing activity levels as well. Dasatinib (BMS-354825, Sprycel®) is a potent multi-target kinase inhibitor of BCR-ABL. Dasatiinib performs better against ABL kinase than earlier drugs such as imatinib, and Dasatinib also is effective against SRC family of kinases, and other receptor kinases such as EPHA2, PDGFR, and c-Kit. Dasatinib's inhibiting potential against Src family kinase members is greater (IC(50) 0.5nM) than its inhibitory activity against ABL (1nM).
