Rhodamine 6G is a highly fluorescent hydrophilic pH-sensitive dye. It is often used as a tracer dye within water to determine the rate and direction of flow and transport. Rhodamine dyes are used extensively in biotechnology applications such as fluorescence microscopy, flow cytometry, fluorescence correlation spectroscopy and ELISA. The dye has a remarkably high photostability and high fluorescence quantum yield. Used as a laser dye and potential mitochondrial probe. The lasing range of the dye is 555 to 585 nm with a maximum at 566 nm. Useful in Pgp efflux assays and has been used to characterize kinetics of MRP1-mediated efflux. The GAT1 gene family includes sodium- and chloride-dependent plasma membrane transporters for neurotransmitters, metabolites and osmolites, which couple substrate flux to transmembrane electrochemical gradients. PROT (Sodium-dependent proline transporter), also known as Solute carrier family 6 member 7, is a 636 amino acid multi-pass membrane protein that is a GAT1 family member specifically expressed in regions of the brain. PROT terminates the action of proline by its high affinity sodium/chloride-dependent reuptake into pre-synaptic terminals. Enriched in glutamatergic synaptic terminals, it is likely that PROT plays an important role in excitatory events of neurotransmission. PROT-mediated proline uptake is inhibited by compounds such as benztropine, LP-403812 and Des-Tyr-Leu-enkephalin (GGFL). These inhibitors of proline uptake may lead to the development of therapeutic agents for certain neurologic disorders.

The four isoforms of potassium/chloride co-transport channels (KCC) belong to a superfamily of cation-chloride co-transporters involved in cell volume maintenance. Nitric oxide (NO) donors activate KCCs, while inhibitors of the cGMP pathway prevent NO donor activation. The ubiquitously expressed KCC1 contains 12 transmembrane domains with both cytoplasmic N and C terminal domains. KCC2 expression is limited to neuronal tissues by a restrictive element similar to the neuronal-restrictive silencing factor. In neurons, KCC2 expression is correlated with an inhibitory response to GABA, while the absence of KCC2 is necessary for an unusual excitatory response to GABA. Alterations of KCC2 expression in the inferior colliculus of rat brain may be related to seizure susceptibility. Conversely, KCC3 is not suspected to play a major role in epilepsy. The two splice variants of KCC3, KCC3a and KCC3b, are predominantly expressed in brain and kidney, respectively, while KCC4 is expressed in muscle, brain, lung, heart and kidney.
