Thermo Scientific Pierce DSP (Lomant's Reagent) is a water-insoluble, homo-bifunctional N-hydroxysuccimide ester (NHS ester) crosslinker that is thiol-cleavable, primary amine-reactive, and useful for many applications. DSP contains an amine-reactive NHS ester at each end of an 8-carbon spacer arm. NHS esters react with primary amines at pH 7–9 to form stable amide bonds and releasing N-hydroxy-succinimide. Proteins, including antibodies, generally have several primary amines in the side chain of lysine (K) residues and the N-terminus of each polypeptide that are available as targets for NHS ester crosslinking reagents. DSP is non-sulfonated and insoluble in water, so it must first be dissolved in an organic solvent and then added to the aqueous reaction mixture. Because DSP does not possess a charged group, it is lipophilic and membrane-permeable and so useful for intracellular and intramembrane conjugation. A sulfonated analog of DSP (DTTSP) is water soluble. DSS, the non-cleavable analog of the DSP crosslinker is also available for applications that require a stable spacer arm that cannot be cleaved in the presence of reducing agents.

The mitochondrial trans-2-enoyl-CoA reductase (MECR), was initially identified as nuclear receptor-binding factor 1 (NRBF1), which can interact with a multitude of nuclear hormone receptors in the presence of the respective ligands. MECR has been shown to be part of the mitochondrial fatty acid synthesis (FAS II) system and to catalyze the NAPDH-dependent reduction of 2-enoyl thioesters, generating saturated acyl-groups. Overexpression of this gene in transgenic mice can lead to cardiac abnormalities, suggesting that inappropriate expression of genes of FAS II can result in the development of hereditary cardiomyopathy.

PDPN, also named as GP36, Gp38, GP40, HT1A-1, OTS8, PA2.26, T1A and T1A-2, has a function in developing testis, most likely at the level of cell-cell interactions among pre-meiotic germ cells and immature Sertoli cells. It may be involved in cell migration and/or actin cytoskeleton organization. When expressed in keratinocytes, PDPN induces changes in cell morphology with transfected cells showing an elongated shape, numerous membrane protrusions, major reorganization of the actin cytoskeleton, increased motility and decreased cell adhesion. It is required for normal lung cell proliferation and alveolus formation at birth. PDPN induces platelet aggregation. It does not have any effect on folic acid or amino acid transport and does not function as a water channel or as a regulator of aquaporin-type water channels. The antibody is special to PDPN.

KLF4 is a member of the Kruppel-like factor (KLF) family of zinc finger transcription factors. Members of this family share 3 contiguous C2H2-type zinc fingers at the carboxyl terminus that comprise the DNA-binding domain. KLF4 is highly expressed in skin and gut epithelial tissues, but is also found in various other cells and tissues, including vascular endothelial cells, lymphocytes, lung, and testis. It is an important regulator of the cell cycle, transcription, and cell differentiation. Together with Sox2, Oct4, and cMyc, KLF4 can induce the reprogramming of primary human fibroblasts to a pluripotent state. KLF4 and other transcription factors can be introduced into cells by DNA transfection, viral infection, or microinjection. Protein transduction using TAT fusion proteins represents an alternative methodology for introducing transcription factors into primary, as well as transformed, cells. Recombinant Human KLF4-TAT is a 483 amino acid protein, including a 13-residue C-terminal TAT peptide, with a calculated molecular weight of 51.7 kDa. PeproTech’s Recombinant Human KLF4-TAT is a mixture of the expected sequence beginning at Met1 and a truncated isoform beginning at Tyr54. Due to post-translational modifications, SDS-PAGE gel shows bands at approximately 72 and 66kDa, under reduced conditions.
