NOTCH4 is a member of the Notch family. Members of this Type 1 transmembrane protein family share structural characteristics including an extracellular domain consisting of multiple epidermal growth factor-like (EGF) repeats, and an intracellular domain consisting of multiple, different domain types. Notch family members play a role in a variety of developmental processes by controlling cell fate decisions. The Notch signaling network is an evolutionarily conserved intercellular signaling pathway which regulates interactions between physically adjacent cells. NOTCH4 is cleaved in the trans-Golgi network, and presented on the cell surface as a heterodimer. NOTCH4 functions as a receptor for membrane bound ligands, and may play a role in vascular, renal and hepatic development. NOTCH4 gene may be associated with susceptibility to schizophrenia in a small portion of cases.

EVER1 Antibody: Epidermodysplasia verruciformis (EV) is an autosomal recessive dermatosis characterized by abnormal susceptibility to human papillomaviruses (HPVs) and a high rate of progression to squamous cell carcinoma on sun-exposed skin. EV is caused by mutations in either of two adjacent genes, EVER1 and EVER2, located on chromosome 17q25.3. Both of these genes encode integral membrane proteins that localize to the endoplasmic reticulum and are predicted to form transmembrane channels. Both EVER1 and EVER2 are members of the transmembrane channel-like (TMC) protein family. EVER1 possesses eight trans-membrane domains and two leucine zipper motifs. EVER1 and EVER2 form a complex and interact with the zinc transporter 1 (ZnT-1), suggesting that EVER1 and EVER2 act to regulate cellular zinc balance.

Description:
Fat Free, also known as ANG2, was identified though a yeast two-hybrid screen using VP53. It is thought to be a structural and functional homolog of the yeast protein VPS51, a protein that together with VPS52, VPS53, and VPS54 form the Golgi-associated retrograde protein (GARP) complex that mediates the tethering and fusion of endosome-derived transport carriers to the trans-Golgi network (TGN). Similar to depletion of any of the VPS52, VPS53, or VPS54, depletion of Fat Free using RNAi impairs protein retrieval to the TGN indicating that Fat Free is missing component of the GARP complex in most eukaryotes.

Lysosome associated membrane protein (LAMP1), also known as lgp120 or lgpA, is a type 1 integral membrane protein that is transported from trans Golgi networks to endosomes and then lysosomes. Upon cell activation, LAMP1 transfer to the plasma membrane is dependent on a carboxyl terminal tyrosine based motif (YXXI). Perturbation in the spacing between the tyrosine based motif relative to the membrane abolishes lysosome localization of LAMP1. This mutant protein then cycles between the plasma membrane and the endosome. Cell surface LAMP1 and LAMP2 have been shown to promote adhesion of human peripheral blood mononuclear cells (PBMC) to vascular endothelium, therefore they are possibly involved in the adhesion of PBMCs to the site of inflammation.

Prominin 2 is a 112 kDa glycoporotein structurally related to Prominin 1 (CD133) although amino acid similarity is not more than 30%, but their genomic organization is strikingly similar. Like Prominin 1, the prominin 2 exhibit similar membrane topology with 5 trans-membrane domains and two large glycosylated extracellular domains. Similar to Prominin1 localization, the Prominin 2 is also associated with membrane protrusions of the epithelial cells from adult kidney, and all along the digestive track and other epithelial tissues.Prominin 2 expression is down-regulated in aggressive prostate cancer cell lines and transient transfection of PROML2 expression vectors has been shown to induce apoptosis in cultured prostate cancer cells, suggesting a tumor suppressive role for Prominin 2. Prominin 2 expression is likely to be involved in growth suppression in the prostate, and down-regulation of Prominin 2 may disrupt normal prostatic homeostasis and lead to uncontrolled prostatic growth.

The Hox proteins play a role in development and cellular differentiation by regulating downstream target genes. Specifically, the Hox proteins direct DNA-protein and protein-protein interactions that assist in determining the morphologic features associated with the anterior-posterior body axis. The mammalian HOX gene complex consists of 39 genes that are located on four linkage groups, which are dispersed over four chromosomes. HOX genes that occupy the same relative position along the 5’ to 3’ coordinate (trans-paralogous genes) are more similar in sequence and expression pattern than adjacent HOX genes on the same chromosome. HoxA3, in conjunction with Pax1, mediates the development of the thymus, parathyroid gland, and carotid body. Members of the suppressor of cytokine signaling (SOCS) family of proteins contain C-terminal regions of homology called the SOCS box, which serves to couple SOCS proteins and their binding partners with the elongin B and C complex. Members of the GGA family, GGA1,GGA2 (also known as VEAR) and GGA3, are multidomain proteins that bind mannose 6-phosphate receptors (MPRs) (1,2,4). GGAs have modular structures with an N-terminal VHS (VPS-27, Hrs, and STAM) domain followed by a GAT (GGA and TOM1) domain, a connecting hinge segment, and a C-terminal GAE (?adaptin ear) domain (5). The amino-terminal VHS domains of GGAs form complexes with the cytoplasmic domains of sorting receptors by recognizing acidic-cluster di-leucine (ACLL) sequences (3). GGA1 and GGA2 do not associate with each other, but they do colocalize on perinuclear membranes (2). The cytosolic domain of memapsin 2, but not that of memapsin 1, binds the VHS domains of GGA1 and GGA2 (6). The human GGA1 gene maps to chromosome 22 and encodes a protein that shares 45% sequence identity with GGA2 and GGA3 (1).
