Description:
Protein phosphatase that is involved in many processes such as microtubule organization at centrosomes, maturation of spliceosomal snRNPs, apoptosis, DNA repair, tumor necrosis factor (TNF)-alpha signaling, activation of c-Jun N-terminal kinase MAPK8, regulation of histone acetylation, DNA damage checkpoint signaling, NF-kappa-B activation and cell migration. The PPP4C-PPP4R1 PP4 complex may play a role in dephosphorylation and regulation of HDAC3. The PPP4C-PPP4R2-PPP4R3A PP4 complex specifically dephosphorylates H2AFX phosphorylated on Ser-140 (gamma-H2AFX) generated during DNA replication and required for DNA double strand break repair. Dephosphorylates NDEL1 at CDK1 phosphorylation sites and negatively regulates CDK1 activity in interphase (By similarity). In response to DNA damage, catalyzes RPA2 dephosphorylation, an essential step for DNA repair since it allows the efficient RPA2-mediated recruitment of RAD51 to chromatin.

Description:
The HUNK (hormonally upregulated Neu-associated kinase) protein, also designated MAK-V in mouse, has been identified as a novel SNF1-related serine/threonine kinase. The human HUNK gene localizes to chromosome 21q22 and encodes a protein with nucleocytoplasmic distribution and localizes to the centrosome. Overexpression of the HUNK protein associates with approximately 50% of breast carcinomas, and may provide diagnostic-prognostic value as a molecular marker. Serine/threonine-protein kinase SNF1-like kinase 2 (SIK) phosphorylates Ser-794 of IRS1 in insulin-stimulated adipocytes, which may modulate the efficiency of insulin signal transduction. SIK is activated by phosphorylation on Thr-175 by STK11 in complex with STE20-related adapter-α and CAB39.

Description:
Cell cycle progression is controlled in part by a family of cyclin proteins and cyclin-dependent kinases (Cdks). Cdk proteins work in concert with the cyclins to phosphorylate key substrates involved in each phase of cell cycle progression. Another family of proteins, Cdk inhibitors, also plays a role in regulating the cell cycle by binding to cyclin-Cdk complexes and modulating their activity. CDKL5 (cyclin-dependent kinase-like 5) is a 1030 amino acid protein that belongs to the CMGC Ser/Thr protein kinase family. Expressed in brain, lung, kidney, prostate, ovary, placenta, pancreas and testis, CDKL5 is thought to play a role in cell cycle regulation. Defects in CDKL5 are a cause of several disorders, such as X-linked infantile spasm syndrome and Rett syndrome.
