KCNK4 is one of the members of the superfamily of potassium channel proteins containing two pore-forming P domains. It homodimerizes and functions as an outwardly rectifying channel. Voltage-gated K+ channels are important determinants of neuronal membrane excitability. Moreover, differences in K+ channel expression patterns and densities contribute to the variations in action potential waveforms and repetitive firing patterns evident in different neuronal cell types (Maletic-Savatic et al., 1995; Pongs, 1999; Blaine and Ribera, 1998; Burger and Ribera, 1996). The Kv3.1 potassium channel is expressed at high levels in neurons that characteristically fire rapid trains of action potentials (Gan et al., 1999). Particularly high levels of this channel are found in neurons of the auditory brainstem. These neurons appear to participate in neural circuits that determine the intensity and timing of auditory stimuli and use this information to determine the location of sounds in space (von Hehn et al., 2004).

Voltage-gated K+ channels in the plasma membrane are important regulators of electrical signaling, controlling the repolarization and the frequency of action potentials in neurons, muscles and other excitable cells. KCNT2 is a 1,135 amino acid multi-pass transmembrane protein belonging to the potassium channel family (calcium-activated subfamily) of proteins. KCNT2 produces rapidly activating outward rectifier potassium currents in reponse to high intracellular sodium and chloride levels. Its channel activity is inhibited by ATP, inhalation anesthetics, such as isoflourane, and upon stimulation of G-protein coupled receptors, such as mAChR M1 and GluR-1. There are four isoforms of KCNT2 that are produced as a result of alternative splicing events.
