The protein encoded by this gene is a type I transmembrane protein that localizes to the endocytic pathway. This protein contains a RING zinc-finger motif and has been shown to possess E3 ubiquitin ligase activity. Expression of this gene in retrovirally transduced T cell hybridoma significantly inhibits activation-induced IL2 and IL4 cytokine production. Induced expression of this gene was observed in anergic CD4(+) T cells, which suggested a role in the induction of anergic phenotype. Alternatively spliced transcript variants encoding distinct isoforms have been reported.

This antibody recognizes a glycoprotein of ~200 kDa, identified as carbonic anhydrase IX (CAIX/gp200). Carbonic Anhydrases (CAs) are members of a large family of zinc metallo-enzymes that catalyze the reversible hydration of carbon dioxide. CAs are involved in a variety of biological processes, including respiration, calcification, acid-base balance, bone resorption and the formation of aqueous humor, cerebrospinal fluid, saliva and gastric juice. They show extensive diversity in distribution and in their subcellular localization. CA IX is specifically expressed in clear-cell renal carcinomas.

KLHL3 protein contains a poxvirus and zinc finger domain at the N-terminus and six tandem repeats (kelch repeats) at the C-terminus. At the amino acid level, KLHL3 shares 77% similarity with Drosophila kelch and 89% similarity with Mayven (KLHL2), another human kelch homolog. At least three isoforms are produced and may be the result of alternative promoter usage. The KLHL3 maps within the smallest commonly deleted segment in myeloid leukemias characterized by a deletion of 5q; however, no inactivating mutations of KLHL3 could be detected in malignant myeloid disorders with loss of 5q.

GATA1 is a Cys2/Cys2 zinc finger DNA binding protein that is expressed primarily in erythroid, megakaryocytic, mast cells and eosinophilic cells. It belongs to the GATA family of transcription factors. GATA1 is a transcriptional activator which probably serves as a general switch factor for erythroid development. It binds to DNA sites with the consensus sequence [AT]GATA[AG] within regulatory regions of globin genes and of other genes expressed in erythroid cells. The protein also plays an important role in erythroid development by regulating the switch from fetal hemoglobin production to adult hemoglobin.

ELAC2 is a zinc phosphodiesterase which displays some tRNA 3' processing endonuclease activity. It is thought to be involved in tRNA maturation, acting by removing a 3' trailer from precursor tRNA. ELAC2 also interacts with activated Smad family member 2 (Smad2) and its nuclear partner forkhead box H1 (also known as FAST-1), and reduced expression can suppress transforming growth factor-beta induced growth arrest. Mutations in this gene result in an increased risk of prostate cancer. A number of allelic variants have been investigated in hereditary prostate cancer.
