Description:
ALKB protects DNA and RNA against damage from methylating compounds from the environment by directly reversing 1-methyladenine (1-meA) and 3-methylcytosine (3-meC) cytotoxic alkylation lesions in DNA and RNA. The enzymes act by oxidative demethylation, utilizing ferrous iron and alpha-ketoglutarate as cofactors, 2-oxoglutarate as a co-substrate, and molecular oxygen as the oxidizing agent. Deficiencies in DNA and RNA repair in mammals are associated with cancer, neurological disease and developmental defects. ALKB plays a role in resistance to anti-cancer drugs which attempt to damage tumor DNA. Escherichia coli ALKB protein belongs to the superfamily of 2-oxoglutarate- and iron(II)-dependent oxygenases.

Description:
Proteins interactions with reactive oxygen agents may result in covalent modifications of amino acid residues in proteins, formation of protein-protein cross-linkages and oxidation of the protein backbone resulting in protein fragmentation. Oxidation targets for Aß(1-16) are the histidine residues coordinated to the metal ions. Copper is bound to Aß in senile plaque of Alzheimer’s disease with Aß (1-16) taking part in the coordination of the Cu2+ ions. Cu2+ and Zn2+ are linked with the neurotoxicity of Aß and free radical damage.

Description:
Pro-inflammatory peptide, acting through G-protein-coupled receptors. Endogenous bradykinin receptor agonist that displays selectivity for B2 over B1 receptors. Endogenously formed following pathophysiological stimuli such as inflammation, tissue damage or anoxia. Activator of endothelial nitric oxide synthase (eNOS; NOS III). Reduces blood pressure by inducing the release of prostacyclin, nitric oxide and endothelium-derived hyperpolarizing factor. Plays an important role in the regulation of fluid and electrolyte balance, non-vascular smooth muscle contraction, vasodilation and capillary permeability.

Description:
Ferritin is a large, iron-storage heteropolymeric protein, which is expressed in most kinds of cells and co-assemble in different proportion in a tissue-specific manner. Ferritin has oligomer of 24 subunits and two types of subunits including light chain(FTL) and heavy chain. Ferritin can remove Fe (â…¡) from solution in the presence of oxygen and is very important for iron homeostasis. Iron is absorbed in the ferrous form and deposited as ferric hydroxides after oxidation. Iron is first oxidized to the ferric state for storage as ferric oxyhdroxide whithin the protein shell of ferritin. Thus, ferritin removes excess iron from the cell sap where it could otherwise participate in peroxidation mechanisms. Ferritin also plays a role in delivery of iron to cells and mediates iron uptake in capsule cells of the developing kidney.

Description:
The protein encoded by this gene belongs to the class-II aminoacyl-tRNA synthetase family. Aminoacyl-tRNA synthetases play critical roles in mRNA translation by charging tRNAs with their cognate amino acids. The encoded protein is a mitochondrial enzyme that specifically aminoacylates alanyl-tRNA. Mutations in this gene are a cause of combined oxidative phosphorylation deficiency 8.
