Description:
SLC29A4 is a member of the equilibrative nucleoside transporter family which plays a key role in nucleoside and nucleobase uptake for salvage pathways of nucleotide synthesis. SLC29A4 is a transmembrane protein that catalyzes the reuptake of monoamines into presynaptic neurons, thus determining the intensity and duration of monoamine neural signaling (1,3). SLC29A4 has been shown to transport several compounds, including serotonin, dopamine, and the neurotoxin 1-methyl-4-phenylpyridinium.

Description:
Members of the BAGE gene family encode antigens that are recognized by cytotoxic T lymphocytes and are also known as CT (cancer/testis) antigens. Generated by juxtacentromeric shuffling of the MLL3 gene, the ancestral BAGE gene was expanded by acrocentric exchanges and/or juxtacentromeric movements.Generally, BAGE proteins are silent in all normal tissues with the exception of testis. BAGE2 and BAGE 3 (B melanoma antigen 2 and 3, respectively), also known as Cancer/testis antigen 2.2 and 2.3 (respectively), are 109 amino acid secreted proteins that are expressed in 22% of melanomas, lung and bladder carcinomas, and are also expressed in normal testis tissue. Like the genes encoding MAGE proteins, BAGE genes are most likely silenced by DNA methylation and/or chromatin compaction in normal tissues other than testis.

Description:
DNA methylation in vertebrate animals is an epigenetic modification that is important for embryonic development, imprinting, and the inactivation of X chromosomes. DNA methylation is catalyzed by a family of DNA methyltransferases (DNMTs) that include the maintenance enzyme DNMT1 and de novo methyltransferases DNMT3a and DNMT3b. The overexpression of DNMT1, DNMT3a, and DNMT3b has been reported in various malignancies, including gastric, urothelial, and lung cancers, and may be related to tumorigenesis, tumor progression, and poor survival. Two isoforms of DNMT3a exist: the full-length DNMT3a, and the shorter form DNMT3a2 which lacks the N-terminal fragment. DNMT3a is expressed ubiquitously at low levels, while DNMT3a2 is specially expressed at high levels in embryonic stem cells and shows restricted expression in tissues known to undergo de novo methylation including testis and ovary. This antibody was raised against the N-terminal region of human DNMT3a. It is expected to detect the 120-130 kDa DNMT3a but not 72-100 kDa DNMT3a2.

Description:
SET07 is a histone methyltransferase that methylates Lys-20 of histone H4. H4 Lys-20 methylation represents a specific tag for epigenetic transcriptional repression. The nuclear SET07 protein, which associates with silent chromatin on euchromatic arms but shows no association with constitutive heterochromatin, prefers nucleosomes as substrate compared to free histones. It appears that SET07 may play a role in maintaining silent chromatin by preventing neighboring acetylation of H4 tail. Although the SET domain contains the active site of enzymatic activity, both sequences upstream and downstream of the SET domain are required for methyltransferase activity.

Description:
DNA methylation, or the addition of methyl groups to cytosine bases in the dinucleotide CpG, is imperative to proper development and regulates gene expression. The methylation pattern involves the enzymatic processes of methylation and demethylation. The demethylation enzyme was recently found to be a mammalian protein, which exhibits demethylase activity associated to a methyl-CpG-binding domain (MBD). The enzyme is able to revert methylated cytosine bases to cytosines within the particular dinucleotide sequence mdCpdG by catalyzing the cleaving of the methyl group as methanol. MeCP2 and MBD1 (PCM1) are first found to repress transcription by binding specifically to methylated DNA. MBD2 and MBD4 (also known as MED1) were later found to colocalize with foci of heavily methylated satellite DNA and believed to mediate the biological functions of the methylation signal. Surprisingly, MBD3 does not bind methylated DNA both in vivo and in vitro. MBD1, MBD2, MBD3, and MBD4 are found to be expressed in somatic tissues, but the expression of MBD1 and MBD2 is reduced or absent in embryonic stem cells, which are known to be deficient in MeCP1 activity. MBD4 have homology to bacterial base excision repair DNA N-glycosylases/lyases. In some microsatellite unstable tumors MBD4 is mutated at an exonic polynucleotide tract.

Description:
LSD1 Antibody: Histone modifications mediate changes in gene expression by altering chromatin structure or by serving as a platform to recruit other proteins. LSD1 is a recently discovered amine oxidase that catalyzes the lysine-specific demethylation of histone proteins via an FAD-dependent oxidative reaction. Methylation on histone H3-K9 is thought to play an important role in heterochromatin formation, while methylation on arginine and some lysine residues (such as H3-K4) is associated with active transcription. LSD1 associates with various proteins, including HDAC1/2, CoREST, and BHC80, that act to regulate LSD1 activity in vivo, and in a histone H3-K4-specific methylase complex that is involved in transcriptional regulation. Experiments have shown that CoREST, a SANT domain-containing corepressor acts to enhance LSD1 activity, while BHC80, a PHD domain-containing protein, inhibits CoREST/LSD1 activity in vitro. LSD1-mediated histone demethylation thus may have significant effects on gene expression.

The N-methyl-D-aspartate receptor 2B, also names as GRIN2B. The sequence of the predicted 1,484-amino acid human protein is 98% and 96% identical to the sequences of the rat and mouse Nmdar2b proteins, respectively. Nmdar2B gene is located on mouse chromosome 6 between Rho and Ly49 centromerically and Glb telomerically. Mapping of the human NMDAR2B receptor subunit gene (GRIN2B) to chromosome 12p12 overexpression of NMDA receptor 2B (NR2B) in the forebrains of transgenic mice leads to enhanced activation of NMDA receptors, facilitating synaptic potentiation in response to stimulation at 10-100 Hz. This antibody is suitable for researchers interested in neuroscience research.
