CDK5 is serine/threonine kinase involved in synaptic regulation and neuronal development; phosphorylates synaptic protein Pctaire1; regulates acetylcholine receptor expression.CDK5 is a member of the cyclindependent kinase family of serine/threonine kinases. It is present in numerous mammalian tissues including kidney, testes, and ovary. Its activity is detected almost exclusively in brain extracts. Neuronal and muscle cells contain the highest amount of this protein. Similar to other Cdks, monomeric Cdk5 displays no enzymatic activity, but Cdk5 is not activated by cyclins. Instead, the p35 protein, which is expressed solely in the brain, activates Cdk5. Cdk5 interacts with D1 and D3 type G1 cyclins and can phosphorylate histone H1, TAU, MAP2 and NF-H and NF-M. Cdk5 activity is involved in terminal differentiation of neurons and muscle cells.

Description:
Interferon regulatory factor 3 (IRF3) is a member of the IRF family. It plays a crucial role in activation of innate immunity and inflammation in response to viral infection, functioning as a molecular switch for antiviral activity. Double-stranded RNA generated during a viral infection leads to IRF3 activation by serine/threonine phosphorylation by TBK1 or IKBKE kinases, which induces a conformational change leading to its dimerization, nuclear localization, and association with CREBBP. The complex, known as DRAF1, activates transcription interferons alpha and beta, as well as other interferon-induced genes. These genes play a critical role in type 1 interferon (IFN)-dependent immune response. A recent study shows phosphorylation of innate immune adaptor proteins MAVS, STING, and TRIF recruits and specifies IRF3 phosphorylation and activation by the serine/threonine-protein kinase TBK1, thereby inducing the production of type-1 interferons.

PDK1 activates conventional PKC and PKC zeta through phosphorylation of critical threonine residues in the activation loop. PDK1 also phosphorylates Protein Kinase B (PKB) at threonine 308 in the presence of phosphatidylinositol-3,4,5-trisphosphate. Active Akt inactivates Glycogen Synthase Kinase 3 (GSK3), eventually leading to the dephosphorylation and activation of glycogen synthase and the stimulation of glycogen synthesis.
