Description:
Calcium-activated and phospholipid-dependent serine/threonine-protein kinase involved in various processes such as regulation of the B-cell receptor (BCR) signalosome, apoptosis and transcription regulation. Plays a key role in B-cell activation and function by regulating BCR-induced NF-κ-B activation and B-cell suvival. Required for recruitment and activation of the IKK kinase to lipid rafts and mediates phosphorylation of CARD11/CARMA1 at 'Ser-559', 'Ser-644' and 'Ser-652', leading to activate the NF-κ-B signaling. Involved in apoptosis following oxidative damage: in case of oxidative conditions, specifically phosphorylates 'Ser-36' of isoform p66Shc of SHC1, leading to mitochondrial accumulation of p66Shc, where p66Shc acts as a reactive oxygen species producer. Acts as a coactivator of androgen receptor (ANDR)-dependent transcription, by being recruited to ANDR target genes and specifically mediating phosphorylation of 'Thr-6' of histone H3 (H3T6ph), a specific tag for epigenetic transcriptional activation that prevents demethylation of histone H3 'Lys-4' (H3K4me) by LSD1/KDM1A. Also involved in triglyceride homeostasis. Serves as the receptor for phorbol esters, a class of tumor promoters.

CXCL16 is a single-pass type I membrane protein, which consists of 246 amino acids, CXCL16 induces a strong chemotatic response and calcium mobilization. CXCL16 acts as a scavenger receptor on macrophages, which specially binds to oxidized low density lipoprotein. CXCL16 may involves in pathophysiology such as atherogenesis. Soluble CXCL16 may play an important role in liver metastases through the induction of epithelial-mesenchymal transition.

Description:
Nitrotyrosine is a marker for inflammation and nitric oxide (NO) production and is formed in the presence of the active metabolite NO. Because nitrotyrosine is a stable product of multiple pathways, such as the formation of peroxynitrite, its plasma concentration may be a useful determinant of NO-dependent damage in vivo. Nitrotyrosine has been detected in inflammatory processes such as septic shock, rheumatoid arthritis, celiac disease, atherosclerotic plaques and chronic renal failure.

Description:
Stores iron in a soluble, non-toxic, readily available form. Important for iron homeostasis. Has ferroxidase activity. Iron is taken up in the ferrous form and deposited as ferric hydroxides after oxidation. Also plays a role in delivery of iron to cells. Mediates iron uptake in capsule cells of the developing kidney (By similarity).

Description:
SUMO4 is a member of the SUMO gene family. This family of small ubiquitin-related modifiers covalently modify target lysines in proteins and control the target proteins' subcellular localization, stability, or activity. Upon oxidative stress, SUMO4 conjugates to various anti-oxidant enzymes, chaperones, and stress defense proteins. This protein may also conjugate to NFKBIA, TFAP2A and FOS, negatively regulating their transcriptional activity, and to NR3C1, positively regulating its transcriptional activity. Covalent attachment to SUMO4 substrates requires prior activation by the E1 complex SAE1-SAE2 and linkage to the E2 enzyme UBE2I. In contrast to SUMO1, SUMO2 and SUMO3, SUMO4 seems to be insensitive to sentrin-specific proteases due to the presence of Pro-90. This may impair processing to mature form and conjugation to substrates. SUMO4 is located in the cytoplasm and specifically modifies IKBA, leading to negative regulation of NF-kappa-B-dependent transcription of the IL12B gene. The M55V substitution has been associated with type I diabetes.

Description:
Guanylate cyclases belong to the adenylyl cyclase class-4/guanylyl cyclase family. There are two forms of guanylate cyclase. The soluble form, known as GCS or sGC, act as receptors for nitric oxide (NO). The membrane-bound receptor form, known as GC, are peptide hormone receptors. GCS is a cGMP-synthesizing enzyme, which is the major receptor for the neurotransmitter nitric oxide. It plays a crucial role in smooth muscle contractility, platelet reactivity and neurotransmission. GCS is a heme-containing heterodimer, consisting of one alpha subunit and one beta subunit. The heme moeity mediates NO activation, and this heme group also binds carbon monoxide (CO), which weakly stimulates the enzyme. Both NO and CO stimulation are enhanced by the allosteric activator 3-(5'-hydroxymethyl-2'furyl)-benzyl-indazole, YC-1. YC-1 can also stimulate GCS in a NO-independent manner. Both alpha and beta subunits are required for cGMP generation, and at least two isoforms exist for each subunit. Heterodimers consisting of alpha-1/beta-1 and alpha-2/beta-1 have been identified, and both display similar enzymatic activity. The distribution of the beta-2 subunit seems to be much more restricted than the beta-1 subunit, with predominant expression in kidney and liver.
