SPP1 Antibody: The secreted protein 1 (SPP1), also known as osteopontin, is a major noncollagenous protein of bone, but is also found in the extracellular matrix of other mineralized tissues and in bodily fluids. In bone, SPP1 is produced by osteoblasts, osteocytes, macrophages, and osteoclasts. SPP1 binds to cells through integrin and non-integrin receptors, as well as the adhesion receptor CD44 in an RGD-independent manner, enabling SPP1 to induce a number of functional effects including macrophage chemotaxis, cytoprotection, and regulation of T helper type 1 cells. Description:
N,N-dimethyl-6-propionyl-2-naphthylamine (prodan) has both an electron-donor and an electron-acceptor substituent, resulting in a large excited-state dipole moment and extensive solvent polarity-dependent fluorescence shifts. When prodan is incorporated into membranes, their fluorescence spectra are sensitive to the physical state of the surrounding phospholipids. In membranes, prodan appears to localize at the surface, although Fourier transform infrared measurements indicate some degree of penetration into the lipid interior. Excited-state relaxation of prodan is sensitive to the nature of the linkage between phospholipid hydrocarbon tails and the glycerol backbone. Tubulin and its hydrophobic surfaces have been probed with the enviroment-sensitive probes prodan. Prodan is also used for the generation of peroxy-oxalate chemiluminescence with H2O2.

Uromodulin (Tamm-Horsfall protein, UMOD) is approx. 85-kDa glycoprotein that is produced in the thick ascending limb of Henle´s loop and early distal convoluted tubules of the nephron. It is a transmembrane protein, which is secreted into the urine through proteolytic cleavage of the glycosylphosphatidylinositol (GPI) anchor. It belongs to the GPI family. Healthy individuals excrete tens of miligrams of uromodulin per day, making in the most abundant protein in the urine. Uromodulin modulates cell adhesion and signal transduction by interacting with cytokines and it inhibits the aggregation of calcium crystals. By reducing calcium oxalate precipitation, uromodulin plays a protective role with respect to renal stone formation as demonstrated by recent studies on THP- deficient mice prone to nephrolithiasis. THP acts as a host defense factor against urinary tract infections induced by uropathogens such as Esherichia coli, Staphylococcus saphrophyticus, Proteus mirabilis and Klebsiela pneumonie. Uromodulin binds to type 1 fimbriae of Escherichia coli and thereby blocks colonization of urothelial cells. Tamm-Horsfall protein interacts with other molecules and cells including IL-1, IL-2, TNF, IgG, neuthrophils, lymphocytes and monocytes. Binding of uromodulin to neutrophils induces synthesis of IL-8, provokes the respiratory burst and degranulation and stimulates chemotaxis and phagocytosis. Recently, genome-wide association studies identified uromodulin as a risk factor for chronic kidney disease and hypertension. Mutations in the Uromodulin gene are associated with three autosomal dominant tubulo-interstitial nephropathies such as familial juvenile hyperuricemic nephropathy (FJHN), medullary cystic kidney disease (MCKD2) and glomerulocystic kidney disease (GCKD). These disorders are characterized by juvenile onset of hyperuricemia, gout and progressive renal failure.

ML RR-S2 CDA is a synthetic cyclic dinucleotide (CDN) that contains non-canonical 2'5'-phosphodiester bonds and is an activator of stimulator of interferon genes (STING).1 It contains mixed linkages (ML) with both 2'5' and 3'5' linkages, which leads to increased thermal stability of human STING in a differential scanning fluorimetry (DSF) assay. ML RR-S2 CDA increases type I interferon production by THP-1 human monocytes relative to unmodified cyclic di-AMP (CDA; Item No. 17753), indicating the ML enhances its action at human STING. It induces expression of IFN-β and the pro-inflammatory cytokines TNF-α, IL-6, and Mcp-1 in murine bone marrow macrophages (BMM) isolated from wild-type, but not STING-/-, mice. ML RR-S2 CDA also induces IFN-β expression in peripheral blood mononuclear cells (PBMCs) isolated from donors carrying STINGWT/WT, STINGWT/REF, and STINGWT/HAQ alleles. In vivo, ML RR-S2 CDA initiates tumor regression and prevents tumor growth upon tumor cell reimplantation in 4T1 breast and CT26 colon cancer mouse xenograft models.
