Description:
ZIP1, also known as ZIRTL (zinc-iron regulated transporter-like), is the first mammalian member of a family of divalent ion transporters. Zinc is an essential ion for cells and plays significant roles in the growth, development, and differentiation. ZIP1 expression is markedly downregulated in a number of cancerous tissues and is thought to function as a tumor suppressor gene in prostate cancer. More recent studies have shown that overexpression of ZIP1 and concomitant increased levels of intracellular zinc in PC-3 cells cause a significant inhibition of NF-kB, leading to down-regulation of the antiapoptotic protein Bcl-2, Bcl-XL, and the apoptosis inhibitor XIAP, reducing the malignant potential of prostate cancer cells in vitro and in vivo.

Description:
Actin reorganization is important for the regulation of neuronal morphology. A protein involved in this process, the transducer of cdc42-dependent actin assembly 1 (TOCA-1) protein, a member of the evolutionarily conserved pombe CDC15 homology (PCH) protein family, is an essential component of the Cdc42 pathway. TOCA-1 binds both N-WASP and Cdc42 and is essential for Cdc42- and PIP2-induced actin polymerization, suggesting that TOCA-1 mediates Cdc42-dependent actin nucleation by activating the N-WASP-WIP complex. Decreased expression of TOCA-1 significantly enhances neurite elongation in PC-12 cells; its overexpression in the same cells suppresses neurite elongation. It has been suggested that TOCA-1 negatively regulates axon branching by regulating membrane trafficking by regulating membrane trafficking through the F-BAR/EFC domain. Multiple isoforms of TOCA-1 are known to exist.

Stanniocalcin 1 (STC1) and stanniocalcin 2 (STC2) are mammalian peptide hormones that were previously considered to be present only in bony fish, where they are involved in calcium homeostasis. STC1 plays a role in calcium and phosphate homoeostasis and is phosphorylated in vitro by protein kinase C, and STC2 is phosphorylated in vitro by casein kinase II (CK2). A human fibrosarcoma cell line, HT1080, expresses both STC1 and STC2 as secreted phosphoproteins in vivo, with STC2 being phosphorylated by an ecto-CK2-like enzyme. STC1 and STC2 have opposite effects on calcium and phosphate homeostasis, namely anti-hypercalcemic and anti-hypocalcemic actions, respectively. STC1 and STC2 are detected in human adrenal tumors, such as pheochromocytoma, differentiated neuroblastoma aldosterone-producing adenoma, and in cultured adrenal tumor cells (rat pheochromocytoma PC-12 cells and human neuroblastoma NB-1 cells).

Expressed in a variety of tissues, ARFRP1 interacts with SYS1 and is thought to be involved in plasma membrane-related signaling events. ARFRP1 exists as multiple alternatively spliced isoforms and is encoded by a gene which maps to a gene cluster on chromosome 20 that is commonly overexpressed in tumors, suggesting a role for ARFRP1 in carcinogenesis.

ARFs are important in eukaryotic vesicular trafficking pathways and they play an essential role in the activation of phospholipase D (PC-PLD). ARL15 (ADP-ribosylation factor-like 15), is a member of the ARF family of the Ras superfamily of small GTPases that are known to be involved in multiple regulatory pathways altered in human carcinogenesis. Other members of this protein family include ARL11, thought to be a tumor suppressor and may play a role in the regulation of apoptosis, ARL2 which plays an important role in microtubule dynamics and cell cycle progression, and ARL4, which may play a role in neurogenesis during embryonic development. The gene encoding ARL15 maps to chromosome 5p15 in humans. At least three isoforms of ARL15 are known to exist.
