The phosphorylation and dephosphorylation of proteins on serine and threonine residues is an essential means of regulating a broad range of cellular functions in eukaryotes, including cell division, homeostasis and apoptosis. A group of proteins that are intimately involved in this process are the serine/threonine (Ser/Thr) protein kinases. STK33 (serine/threonine kinase 33) is a 514 amino acid protein that belongs to the CaMK (calcium/calmodulin dependent kinase) subfamily of structurally related serine/threonine kinases. Widely expressed at low levels with predominant expression in testis, lung, retina and fetal organs such as brain, heart and spinal cord, STK33 contains one protein kinase domain and functions as a Ser/Thr protein kinase with a possible role in spermatogenesis. The gene encoding STK33 lies within a region on chromosome 11 that has been associated with a variety of defects, including Long QT syndrome, T-cell leukemia, Beckwith-Wiedemann syndrome, Usher syndrome 1C and various other malignancies.

TSSK 6 (testis-specific serine kinase 6), also known as SSTK, TSSK4, FKSG82 or CT72, is a 273 amino acid protein that contains one protein kinase domain and belongs to the Ser/Thr protein kinase family. Highly expressed in testis with lower expression in ovary, colon, thymus, small intestine and spleen, TSSK 6 catalyzes the ATP-dependent phosphorylation of proteins involved in sperm production and chromatin remodeling. TSSK 6 uses magnesium as a cofactor and is thought to be required for proper sperm development and function, as well as DNA condensation events. Defects in the gene encoding TSSK 6 are associated with male infertility characterized by low sperm count and decreased sperm motility.

The tumor suppressor PTEN plays an essential role in regulating signaling pathways involved in cell growth and apoptosis and is inactivated in a wide variety of tumors. Protein interacting with PTEN carboxyl terminus 1 (PICT-1), also designated p60 or Glioma tumor suppressor candidate region gene 2 protein, binds to the C-terminus of PTEN and regulates its turnover. Five Ser/Thr residues within the C-terminal segment of PTEN, including Ser-380, are phosphorylated upon binding of PTEN to PICT-1 and may contribute to the stabilization of PTEN. PICT-1 is localized to the nucleus and/or nucleolus and is highly expressed in pancreas and heart, but can also be detected in liver, skeletal muscle, placenta and kidney. PICT-1 also interacts with herpes simplex virus 1 regulatory proteins ICP22 and ICP0. The tumor suppressor GLTSCR2 gene, which encodes PICT-1, is located in a 150-kb minimal common deletion region for human gliomas, especially oligodendrogliomas, and maps to human chromosome 19q13.3.
