Hepatocyte Cell Adhesion Molecule (HEPACAM) is a single-pass type I membrane protein that localizes to the cytoplasmic side of the cell membrane. HEPACAM includes a signal sequence (amino acid 1-33), an extracellular region (amino acid 34-240) with one Ig-like C2-type domain and one Ig-like V-type domain, a transmembrane segment (amino acid 241-261), and a cytoplasmic domain (amino acid 262 - 416). The cytoplasmic domain plays an important role in regulation of cell-matrix adhesion and cell motility. HEPACAM acts as a homodimer and dimer formation occurs predominantly through cis interactions on the cell surface. HEPACAM is involved in cell motility and cell-matrix interactions. The expression of this gene is down-regulated or undetectable in many cancer cell lines, so this may be a tumor suppressor gene.

CD33 is a 67 kDa type I transmembrane glycoprotein and a member of the sialoadhesin family of cell surface receptors. It is absent from pluripotent stem cells but appears on myelomonocytic precursors after CD34. It then continues to be expressed on both the myeloid and monocyte lineages. While it has been reported that CD33 can function as a sialic acid-dependent cell adhesion molecule, cells expressing CD33 require desialylation before they can bind cells bearing the appropriate sialoglycoconjugates. This suggests that inhibitory cis interactions may regulate or block any adhesion function.

The Semphorins are a large family of phylogenetically conserved proteins that play a pivotal role in maintaining homeostasis in the immune system. Twenty members of this family have been identified and categorized into eight subclasses based on sequence similarity and distinctive structural features. CD100, also known as Sema4D, is a 150 kDa transmembrane class IV semaphorin. Studies have shown that CD100 can induce monocyte migration, T-cell activation, and B-cell survival, as well as T/B cell and T/DC “cooperation”. The CD100 precursor contains 862 amino acids, including a 21 a.a. signal sequence, a 713 a.a. extracellular domain, a 21 a.a. transmembrane sequence, and a 107 a.a. cytoplasmic region.
