Description:   The MP6-XT22 monoclonal antibody specifically reacts with mouse tumor necrosis factor alpha (TNF-alpha), a 156 amino pro-inflammatory cytokine. It is secreted by various cells including adipocytes, activated monocytes, macrophages, B cells, T cells and fibroblasts. NF-α is cytotoxic to a wide variety of tumor cells and is an essential factor in mediating the immune response against bacterial infections. TNF-α also plays a role in the induction of septic shock, auto immune diseases, rheumatoid arthritis, inflammation, and diabetes. The MP6-XT22 is reported to neutralize the bioactivity of natural or recombinant TNF-alpha and is useful for intracellular immunofluorescent staining.

Description:   The 145-2C11 monoclonal antibody reacts with mouse CD3e, the 20 kDa ε chain of the TCR complex. Together with the γ and δ subunits of CD3, the ε subunits are involved in the assembly, trafficking, and surface expression of T-cell receptor complex. CD3 is expressed on thymocytes, mature T cells, and natural killer T cells, and the ε chain enhances the antigen recognition. The 145-2C11 antibody binds to the TCR complex and, depending on the conditions, initiates T cells activation, proliferation, and apoptosis. The soluble antibody seems to block lysis of target cells by antigen-specific cytotoxic T lymphocytes. The 145-2C11 antibody does not cross-react with the rat leukocytes, and it is used as a phenotypic marker for mouse T lymphocytes.

Description:   The OKT4 monoclonal antibody specifically binds to the CD4 receptor for the human immunodeficiency virus (HIV). CD4 is a 59 kDa single-chain transmembrane glycoprotein that expressed on the surface of most of the thymocytes, T-helper cells, and in low levels on monocytes and macrophages. CD4 is a co-receptor in the antigen-induced T cell activation (together with the MHC class II). The OKT4 and the RPA-T4 monoclonal antibodies recognize different epitopes of CD4 and they do not exhibit cross-block binding.BG Violet 450 conjugate is an alternative to the Pacific Blue, eFluor 450, or BD Horizon V450 dyes. It is excited by the violet (405 nm) laser, with a peak emission of 450nm.

Description:   The RM4-5 monoclonal antibody specifically reacts with mouse CD4, also known as L3T4, a 55 kDa differentiation antigen expressed by the majority of thymocytes, subpopulations of mature T cells (like major histocompatibility complex class II-restricted T lymphocytes), a subset of natural killer T cells, and on pluripotent hematopoietic stem cells. CD4 binds to the major histocompatibility complex class II (MHC class II) and enhances T lymphocyte development and mature T cells functions. In T lymphocytes, CD4 binds to the cytoplasmic tail of enzyme tyrosine kinase (p56lck).Binding of RM4-5 is blocked by the anti-mouse CD4 clone GK1.5.

Description:   The RB6-8C5 monoclonal antibody reacts with the mouse Ly-6G (also known as Gr-1). The Ly-6G protein is a myeloid differentiation antigen of 21-25 kDa, expressed in a regulated manner by the myeloid lineage in the bone marrow, where the level of antigen expression is correlated with the granulocyte maturation and differentiation. In the bone marrow, the antigen is not expressed by the erythroid cells. From the peripheric cells, RB6-8C5 binds with monocytes, neutrophils and eosinophils. As a marker for the mouse monocytes, macrophages and granulocytes, the RB6-8C5 antibody is usually combined with M1/70, a macrophage labeling antibody (Anti-CD11b), for phenotypic analysis.

Description:   The BM8.1 monoclonal antibody specifically binds to the Mouse125 kDa F4/80 antigen, expressed by most mature macrophages. F4/80 is a transmembrane protein used as a marker of macrophages, although it is also expressed on Kupffer and Langerhans cells. The expression of F4/80 antigen is upregulated on bone marrow cells stimulated in vitro with the macrophage colony stimulating factor. The F4/80 antigen is a requirement for the induction of CD8 T cells-mediated peripheral tolerance.BG Violet 450 conjugate is an alternative to the Pacific Blue, eFluor 450, or BD Horizon V450 dyes. It is excited by the violet (405 nm) laser, with a peak emission of 450nm.

Description:   Tricine was first prepared by Good for use as a buffer for chloroplast reactions. It is structurally similar to Tris, but is much less inhibitory at high concentrations. For ATP assays using firefly luciferase, tricine buffer at 25 mM was found to be the best of ten common buffers tested.
Tricine is used as buffer component for separation of low molecular weight peptides. Tricine can be used in cryopreservation medium for the preservation of tissues and organs. Cryopreservation depends on the physical and chemical characteristics of the preservation medium used. The pH values and pK values for tricine/DMSO mixtures has been reported down to -20 °C. Tricine has been found to be an efficient scavenger of hydroxyl radicals in a study of radiation-induced membrane damage. Tricine is typically the buffer of choice in SDS-PAGE systems when separating proteins in the range of 1 to 100 kDa.
A buffer may be prepared by titrating with sodium hydroxide to the desired pH, using about a half-equivalent of NaOH.

Description:   The 2.43 monoclonal antibody specifically reacts with mouse CD8 antigen. CD8a (the alpha chains) form heterodimers with CD8b (the beta chains) or homodimers (alpha-alpha), which occur as receptors on the surface of the majority of thymocytes. A subpopulation of mature T lymphocytes expresses the CD8 alpha beta (alpha beta TCR T cells), and a subpopulation of intestinal intraepithelial lymphocytes and dendritic cells express CD8a without CD8b. CD8 interacts with the mouse major histocompatibility complex class I (MHC class I) molecules on antigen-presenting cells or epithelial cells.

Description:   The 30-H12 monoclonal antibody specifically binds to MouseCD90.2, an alloantigen known as Thy-1.2, expressed on thymocytes, mature T cells, epithelial cells, neurons, hematopoietic stem cells, and fibroblasts. CD90 is a membrane glycoprotein that regulates the adhesion and signal transduction in T lymphocytes, and the adhesion of thymocytes to thymic stroma. The interaction between 30-H12 and the antibody to the CD3/TCR complex upregulates thymocytes signal transduction and apoptosis and downregulates mature T cell proliferation. The 30-H12 antibody seems to be unable to cross-link with CD90.1.

Description:   The UCHT1 monoclonal antibody specifically reacts with the ε chain of the CD3/T lymphocyte antigen receptor complex. The CD3 complex contains γ, δ, and ε chains, and it is part of the TCR complex, expressed by all mature T lymphocytes and by the thymocyte lineage. CD3 enhances the antigen recognition by signal transduction. Unlike HIT3a, another specific antibody of CD3, the UCHT1 antibody can stain both the surface and intracellular CD3ε. The immobilized UCHT1 can cross-link with the TCR complex, enhancing cellular activation and proliferation.

Description:   The ACK2 monoclonal antibody specifically reacts with mouse CD117 (c-Kit receptor), a 145 kDa transmembrane tyrosine-kinase receptor encoded by the Kit gene. The c-Kit receptor, also known as stem cell factor receptor, is expressed on hematopoietic progenitor cells in adult bone marrow, in progenitors of erythroid and myeloid lineages, and precursors of B and T cells. CD117 enhances the proliferation and the differentiation of the hematopoietic progenitor cells and seems to enhance the development of T cells, as the c-Kit receptor and its ligand are expressed by the thymus.
