In eukaryotes, the phosphorylation and dephosphorylation of proteins on serine and threonine residues is an essential means of regulating a broad range of cellular functions, including division, homeostasis and apoptosis. A group of proteins that are intimately involved in this process are the protein phosphatases. In general, the protein phosphatase (PP) holoenzyme is a trimeric complex composed of a regulatory subunit, a variable subunit, and a catalytic subunit. Four major families of protein phosphatase catalytic subunits have been identified, designated PP1, PP2A, PP2B (calcineurin) and PP2C. An additional protein phosphatase catalytic subunit, PPX (also known as PP4) is a putative member of a novel PP family. The PP2A family comprises subfamily members PP2A Alpha and PP2A Beta. The PP2A catalytic subunit associates with a variety of regulatory subunits. Regulatory subunits include PP2A-A-Alpha and -A-Beta, PP2A-B-Alpha and -B-Beta, PP2A-C-Alpha and -C-Beta, PP2A-B56-Alpha, -B56-Beta, -B56-gamma and -B56-Delta.

This antibody recognizes a protein of 12 kDa, identified as β2-microglobulin. Major histocompatibility complex (MHC) class 1 molecules bind to antigens for presentation on the surface of cells. The proteasome is responsible for producing these antigens from the components of foreign pathogens. MHC class 1 molecules consist of analpha heavy chain that contains three subdomains (α1,alpha2,alpha3) and a non-covalent associating light chain, known as beta-2-Microglobulin. beta-2-Microglobulin associates with thealpha3 subdomain of thealpha heavy chain and forms an immunoglobulin domain-like structure that mediates proper folding and expression of MHC class 1 molecules. Thealpha1 andalpha2 domains of thealpha heavy chain form the peptide antigen-binding cleft. Mutations in the beta-2-Microglobulin gene can enhance the progression of malignant melanoma phenotypes.

Glucose-6-phosphatase (G6Pase), is a multicomponent enzyme system that hydrolyzes glucose-6-phosphate (G6P) in the final step of gluconeogenesis and gluconeolysis. G6Pase localizes to the endoplasmic reticulum, and while liver, kidney, and intestine are the only tissues that express the first identified isoform, G6Pase-Alpha, a second form, designated G6Pase-Beta, contributes to blood glucose homeostasis in a wider range of tissues. G6Pase-Beta, also known as SCN4, UGRP or G6PC3 (glucose 6 phosphatase, catalytic, 3), is a 346 amino acid endoplasmic reticulum multi-pass membrane protein that is involved in carbohydrate biosynthesis and the gluconeogenesis pathway. Inhibited by vanadate, G6Pase-Beta hydrolyzes GP6 to glucose in the endoplasmic reticulum. Due to its necessary involvement in normal glucose metabolism, G6Pase-Beta may play an integral role in diabetes and glycogen storage diseases (GSDs).
