Description:
GAPVD1 is a 1478 amino acid peripheral membrane protein that acts both as a GTPase-activating protein (GAP) and a guanine nucleotide exchange factor (GEF). GAPVD1 participates in many processes such as insulin receptor internalization, Glut4 trafficking and endocytosis. In addition, depletion of GAPVD1 leads to delayed EGFR degradation by mediating receptor ubiquination through its RGD domain, suggesting that it may be an important mediator of carcinogenesis resulting from Ras protein mutations. There are six isoforms of GAPVD1 that are produced as a result of alternative splicing events.

Description:
The protein encoded by this gene is a guanine nucleotide exchange factor (GEF) similar to the Saccharomyces cerevisiae CDC25 gene product. Functional analysis has demonstrated that this protein stimulates the dissociation of GDP from RAS protein. The studies of the similar gene in mouse suggested that the Ras-GEF activity of this protein in brain can be activated by Ca2+ influx, muscarinic receptors, and G protein beta-gamma subunit. Mouse studies also indicated that the Ras-GEF signaling pathway mediated by this protein may be important for long-term memory. Alternatively spliced transcript variants encoding distinct isoforms have been reported. [provided by RefSeq, Mar 2009]

Description:
ARHGEF7 is a p21-activated protein kinase (Pak)-interacting exchange factor that has been identified as a putative guanine nucleotide exchange factor (GEF) for Rac/Cdc42. It is expressed ubiquitously in all the tissue examined and other isoforms are expressed mainly in the central nervous system. Regulated phosphorylation of ARHGEF7 is necessary to maintain the balance between normal signalling by EGFR and Src versus aberrant growth and transformation.Multiple alternatively spliced transcript variants encoding different isoforms have been described for this gene, but some of their full-length sequences have not been determined.

Description:
This gene is a member of the VAV gene family. The VAV proteins are guanine nucleotide exchange factors (GEFs) for Rho family GTPases that activate pathways leading to actin cytoskeletal rearrangements and transcriptional alterations. The encoded protein is important in hematopoiesis, playing a role in T-cell and B-cell development and activation. The encoded protein has been identified as the specific binding partner of Nef proteins from HIV-1. Coexpression and binding of these partners initiates profound morphological changes, cytoskeletal rearrangements and the JNK/SAPK signaling cascade, leading to increased levels of viral transcription and replication. Alternatively spliced transcript variants encoding multiple isoforms have been observed for this gene. [provided by RefSeq, Apr 2012].

Description:
This gene encodes a member of the human ARF gene family, which is part of the RAS superfamily. The ARF genes encode small guanine nucleotide-binding proteins that stimulate the ADP-ribosyltransferase activity of cholera toxin and play a role in vesicular trafficking and as activators of phospholipase D. The product of this gene is localized to the plasma membrane, and regulates vesicular trafficking, remodelling of membrane lipids, and signaling pathways that lead to actin remodeling. A pseudogene of this gene is located on chromosome 7.

Description:
N-RAS Antibody: Activating mutations and overexpression of classical Ras subfamily members (N-RAS, H-RAS, and K-RAS) have been widely investigated as key events in the development of human cancers. The N-RAS protein shuttles between the Golgi apparatus and the plasma membrane. This shuttling is regulated through palmitoylation and depalmitoylation by the ZDHHC9-GOLGA7 complex. N-RAS, which has intrinsic GTPase activity, is activated by a guanine nucleotide-exchange factor and inactivated by a GTPase activating protein. Mutations in this gene have been associated with somatic rectal cancer, follicular thyroid cancer, autoimmune lymphoproliferative syndrome, Noonan syndrome, and juvenile myelomonocytic leukemia.

Description:
Orphan receptor tyrosine kinase (RTK) that plays a role in epithelial cell differentiation and regionalization of the proximal epididymal epithelium. May activate several downstream signaling pathways related to cell differentiation, proliferation, growth and survival including the PI3 kinase-mTOR signaling pathway. Mediates the phosphorylation of PTPN11, an activator of this pathway. May also phosphorylate and activate the transcription factor STAT3 to control anchorage-independent cell growth. Mediates the phosphorylation and the activation of VAV3, a guanine nucleotide exchange factor regulating cell morphology. May activate other downstream signaling proteins including AKT1, MAPK1, MAPK3, IRS1 and PLCG2.
