Description:
WDFY3 was identified in a screen for phosphatidylinositol 3 phosphate binding proteins. The FYVE domain is a zinc binding domain found in many PtdIns binding proteins and is proposed to target proteins to membrane lipids. WDFY3 has been shown localized to the nuclear membrane and within autophagic membranes and it is proposed to function as a link between protein aggregates and the autophagic machinery. WDFY3 is also known as autophagy linked FYVE protein.

Description:
Sphingosine-1-phosphate (SPP) is a novel lipid messenger with both intracellular and extracellular functions. Intracellularly, it regulates proliferation and survival, and extracellularly, it is a ligand for EDG1 (MIM 601974). Various stimuli increase cellular levels of SPP by activation of sphingosine kinase (SPHK), the enzyme that catalyzes the phosphorylation of sphingosine. Competitive inhibitors of SPHK block formation of SPP and selectively inhibit cellular proliferation induced by a variety of factors, including platelet-derived growth factor (e.g., MIM 173430) and serum.[supplied by OMIM].

Description:
Regulates RHOA activity, and plays a role in cytoskeleton remodeling. Necessary for normal completion of cytokinesis. Plays a role in maintaining normal diacylglycerol levels in the Golgi apparatus. Binds phosphatidyl inositol phosphates (<i>in vitro</i>). May catalyze the transfer of phosphatidylinositol and phosphatidylcholine between membranes (By similarity). Necessary for maintaining the normal structure of the endoplasmic reticulum and the Golgi apparatus. Required for protein export from the endoplasmic reticulum and the Golgi. Binds calcium ions.

Description:
CD45R, also known as B220, a member of the protein tyrosine phosphate family and a major cell surface glycoprotein, represents a restricted form of the CD45 family which primarily recognizes only cells of B-lineage from pro-B cell through mature B lymphocytes and prior to the availability of anti-CD19 monoclonal antibodies was commonly used as a pan B-cell marker. It also reacts with certain activated T cells as well as non-MHC-restricted lytically active lymphokine-activated killer (LAK) cells. In vivo administration of RA3-6B2 has been shown to affect differentiation of both T and B cells in normal mice and reduce the level of anti-DNA antibodies and lymphadenopathies in MRL/lpr mice.

Description:
Multifunctional protein that plays a central role in the cellular response to oxidative stress. The two major activities of APEX1 in DNA repair and redox regulation of transcriptional factors. Functions as a apurinic/apyrimidinic (AP) endodeoxyribonuclease in the DNA base excision repair (BER) pathway of DNA lesions induced by oxidative and alkylating agents. Initiates repair of AP sites in DNA by catalyzing hydrolytic incision of the phosphodiester backbone immediately adjacent to the damage, generating a single-strand break with 5'-deoxyribose phosphate and 3'-hydroxyl ends. Does also incise at AP sites in the DNA strand of DNA/RNA hybrids, single-stranded DNA regions of R-loop structures, and single-stranded RNA molecules. Has a 3'-5' exoribonuclease activity on mismatched deoxyribonucleotides at the 3' termini of nicked or gapped DNA molecules during short-patch BER. Possesses a DNA 3' phosphodiesterase activity capable of removing lesions (such as phosphoglycolate) blocking the 3' side of DNA strand breaks. May also play a role in the epigenetic regulation of gene expression by participating in DNA demethylation. Acts as a loading factor for POLB onto non-incised AP sites in DNA and stimulates the 5'-terminal deoxyribose 5'-phosphate (dRp) excision activity of POLB. Plays a role in the protection from granzymes-mediated cellular repair leading to cell death. Also involved in the DNA cleavage step of class switch recombination (CSR). On the other hand, APEX1 also exerts reversible nuclear redox activity to regulate DNA binding affinity and transcriptional activity of transcriptional factors by controlling the redox status of their DNA-binding domain, such as the FOS/JUN AP-1 complex after exposure to IR. Involved in calcium-dependent down-regulation of parathyroid hormone (PTH) expression by binding to negative calcium response elements (nCaREs).

Description:
SPHK(Sphingosine kinase 1) is also named as SPHK, SPK.It catalyzes the phosphorylation of sphingosine to form sphingosine 1-phosphate (SPP), a lipid mediator with both intra- and extracellular functions and is precariously perched at the balance point between progrowth and pro-death signalling in the cell.Endogenous SPHK1 purified from placenta has identical enzymatic characteristics, suggesting posttranslational modification does not effect functional properties. It has 2 isoforms produced by alternative splicing and can form the oligomeric structure.

Description:
Glucosamine 6-phosphate N-acetyltransferase (GNA1), also designated phosphoglucosamine transacetylase or phosphoglucosamine acetylase, belongs to the GNA1 subfamily of the larger acetyltransferase family of proteins. GNA1, a peripheral membrane protein containing one N-acetyltransferase domain, is expressed in the colon and maps to cytoband 14q22.1. The protein localizes to the Golgi apparatus and the endosome. It is important for UDP-GlcNAc biosynthesis pathway. GNA1 catalyzes the synthesis of GlcNAc6P from AcCoA and GlcN6P, a step in the UDP-GlcNAc6P formation pathway.
