EGFLAM is a 1,017 amino acid secreted protein containing three EGF-like domains, two fibronectin type-III domains, and three laminin G-like domains. Colocalizing with bassoon, CtBP and dystroglycan in photoreceptor synaptic terminals, EGFLAM is involved in retinal photoreceptor ribbon synapse formation. EGFLAM may also promote matrix assembly and cell adhesion. Existing as five alternatively spliced isoforms, the gene encoding EGFLAM maps to human chromosome 5p13.2. Chromosome 5 makes up approximately 6% of the human genome and contains 181 million base pairs, which encode 1,000 genes. Cockayne syndrome, Treacher Collins syndrome, acute myelogenous leukemias and myelodysplastic syndrome are associated with genes present on chromosome 5.

FNDC1 is moderately expressed in skeletal muscle, pancreas, heart, kidney, spinal cord, ovary and lung. Expression of FNDC1 is induced in response to hypoxia in ventricular cardiomyocytes. Since FNDC1 interacts with G∫ and G©, it is likely that FNDC1 is an activator for G-protein signaling. Though normally absent in healthy skin, FNDC1 expression is induced by TGF-beta signaling in skin tumors and psoriasis. There are two isoforms of FNDC1 that are produced as a result of alternative splicing events.

Phosphatase enzymes catalyse hydrolysis of phosphoric acid esters of various alcohols, e.g. a hexose phosphate, to yield the alcohol and free inorganic phosphate. This may be a way of recycling phosphate in the cell, and the level of phosphate may be elevated under conditions of phosphate starvation (in algae for example). FNDC3A is a 1,134 amino acid protein that belongs to the FNDC3 family of proteins. FNDC3A contains an N-terminal proline-rich region, nine fibronectin type-III domains (none of which contain an RGD sequence) and a hydrophobic C-terminal transmembranous helix. Expressed in a wide variety of tissues, FNDC3A localizes to Golgi vesicles and to the developing acrosome of spermatids. FNDC3A is believed to function in glycosaminoglycan and collagen synthesis. In mice, a mutation in the gene encoding FNDC3A causes male sterility due to defective adhesion between Sertoli cells and spermatids in the seminiferous epithelium. This suggests that FNDC3A plays an important role in spermatogenesis, possibly mediating or maintaining the adhesion between Sertoli cells and spermatids.

The cystatin superfamily encompasses proteins that contain multiple cystatin-like sequences. Some of the members are active cysteine protease inhibitors, while others have lost or perhaps never acquired this inhibitory activity. There are three inhibitory families in the superfamily, including the type 1 cystatins(stefins), type 2 cystatins and the kininogens. The type 2 cystatin proteins are a class of cysteine proteinase inhibitors found in a variety of human fluids and secretions. The cystatin locus on chromosome 20 contains the majority of the type 2 cystatin genes and pseudogenes. This gene is located in the cystatin locus and encodes a type 2 salivary cysteine peptidase inhibitor. The protein is an S-type cystatin, based on its high level of expression in saliva, tears and seminal plasma. The specific role in these fluids is unclear but antibacterial and antiviral activity is present, consistent with a protective function.
