Description:
Transport of phosphorylated lysosomal enzymes from the Golgi complex and the cell surface to lysosomes. Lysosomal enzymes bearing phosphomannosyl residues bind specifically to mannose-6-phosphate receptors in the Golgi apparatus and the resulting receptor-ligand complex is transported to an acidic prelyosomal compartment where the low pH mediates the dissociation of the complex. This receptor also binds IGF2. Acts as a positive regulator of T-cell coactivation, by binding DPP4.

Description:
Sphingosine kinase (SPHK) phosphorylates sphingosine to form a bioactive lipid mediate, sphingosine 1-phosphate (S1P or SPP). S1P is a novel lipid messenger with both intracellular and extracellular functions. Intracellularly, it regulates proliferation and survival while extracellularly it is a ligand for EDG 1. Expression of SPHK1 and subsequent production of S1P promotes the G1-S transition, protects cells from apoptosis, stimulates migration and angiogenesis, and induces tumor formation in mice.

Description:
Key component of innate and adaptive immunity. TLRs (Toll-like receptors) control host immune response against pathogens through recognition of molecular patterns specific to microorganisms. TLR9 is a nucleotide-sensing TLR which is activated by unmethylated cytidine-phosphate-guanosine (CpG) dinucleotides. Acts via MYD88 and TRAF6, leading to NF-kappa-B activation, cytokine secretion and the inflammatory response. Controls lymphocyte response to Helicobacter infection.

Description:
CD45R, also known as B220, a member of the protein tyrosine phosphate family and a major cell surface glycoprotein, represents a restricted form of the CD45 family which primarily recognizes only cells of B-lineage from pro-B cell through mature B lymphocytes and prior to the availability of anti-CD19 monoclonal antibodies was commonly used as a pan B-cell marker. It also reacts with certain activated T cells as well as non-MHC-restricted lytically active lymphokine-activated killer (LAK) cells. In vivo administration of RA3-6B2 has been shown to affect differentiation of both T and B cells in normal mice and reduce the level of anti-DNA antibodies and lymphadenopathies in MRL/lpr mice.

Description:
Potent and selective inhibitor of heme oxygenase (Ki=3nM), the enzyme that generates carbon monoxide (CO). Inhibits soluble guanylyl cyclase. Produces a time- and concentration-dependent inactivation of all three isoforms of nitric oxide synthase (IC50=0.8µM, 4.0µM, and 5.0µM for nNOS (NOS I), iNOS (NOS II) and eNOS (NOS III), respectively). Does not cross the blood-brain barrier. An useful tool to elucidate the role of CO in signal transduction and as a neurotransmitter.

Description:
Sphingosine-1-phosphate (SPP) is a novel lipid messenger with both intracellular and extracellular functions. Intracellularly, it regulates proliferation and survival, and extracellularly, it is a ligand for EDG1 (MIM 601974). Various stimuli increase cellular levels of SPP by activation of sphingosine kinase (SPHK), the enzyme that catalyzes the phosphorylation of sphingosine. Competitive inhibitors of SPHK block formation of SPP and selectively inhibit cellular proliferation induced by a variety of factors, including platelet-derived growth factor (e.g., MIM 173430) and serum.[supplied by OMIM].

CPOX(Coproporphyrinogen-III oxidase, mitochondrial) is also named as CPO and CPX. Human CPO is a 76 kDa protein that is active as a homodimer in the mitochondrial intermembrane pace.Mutations in human CPO gene predict the clinical outcome of the disease, with either hepatic hereditary coproporphyria or hematological manifestations of erythropoietic harderoporphyria.CPOX has a transit peptide of 110 aa and the chain contains 344 aa.

Description:
Component of triglyceride-rich very low density lipoproteins (VLDL) and high density lipoproteins (HDL) in plasma. Plays a multifaceted role in triglyceride homeostasis. Intracellularly, promotes hepatic very low density lipoprotein 1 (VLDL1) assembly and secretion; extracellularly, attenuates hydrolysis and clearance of triglyceride-rich lipoproteins (TRLs). Impairs the lipolysis of TRLs by inhibiting lipoprotein lipase and the hepatic uptake of TRLs by remnant receptors.
