Transaldolase 1 is a key enzyme of the nonoxidative pentose phosphate pathway providing ribose-5-phosphate for nucleic acid synthesis and NADPH for lipid biosynthesis. This pathway can also maintain glutathione at a reduced state and thus protect sulfhydryl groups and cellular integrity from oxygen radicals. The functional gene of transaldolase 1 is located on chromosome 11 and a pseudogene is identified on chromosome 1 but there are conflicting map locations. The second and third exon of this gene were developed by insertion of a retrotransposable element. This gene is thought to be involved in multiple sclerosis.

STK31 (Serine/threonine protein kinase 31) is similar to a mouse gene that encodes a putative protein kinase with a tudor domain, and shows testis specific expression. Alternative splicing results in multiple transcript variants encoding different isoforms. Protein kinases are enzymes that transfer a phosphate group from a phosphate donor, generally the g phosphate of ATP, onto an acceptor amino acid in a substrate protein. By this basic mechanism, protein kinases mediate most of the signal transduction in eukaryotic cells, regulating cellular metabolism, transcription, cell cycle progression, cytoskeletal rearrangement and cell movement, apoptosis, and differentiation. The PHEX a 749 amino acid protein that putatively consists of an intracellular, transmembrane and extracellular domain. PHEX mutations have been observed in 60-80% of hypophosphatemic rickets patients. The PHEX protein, which is a single-pass membrane protein, is also designated HYP, X-linked hypophosphatemia protein or metalloendopeptidase homolog PEX. PHEX plays an active role in bone and dentin mineralization and renal phosphate re-absorption. X-linked hypophosphatemic rickets, also designated HYP, is an X-linked dominant disorder characterized by impaired phosphate uptake in the kidney, which is likely to be caused by abnormal regulation of sodium phosphate cotransport in the proximal tubules. Clinical manifestations include skeletal deformities, growth failure, craniosynostosis, paravertebral calcifications, pseudofractures in lower extremities, and muscular hypotonia with onset in early childhood.

During fertilization in mammals, the sperm activates the egg by causing an increase in the level of free cytoplasmic calcium concentration. This increased calcium concentration induces a characteristic series of oscillations that trigger egg activation and early embryo development. A hamster protein named oscillin is thought to be involved in this pathway. The enzyme glucosamine-6-phosphate isomerase (GNPI) or deaminase (GNPDA1) and the related protein GNPDA2 are the human homologs of hamster oscillin. GNPDA1 and GNPDA2 catalyze the conversion of GNP to fructose-6-phosphate and ammonia. Both proteins exist as homohexamers and are ubiquitously expressed with highest expression in testis, ovary and heart. Three isoforms of GNPDA2 are expressed due to alternative splicing events.

Tubulin is the major constituent of microtubules, existing as a heterodimer of the alpha and beta subunits. The beta III isoform of tubulin is found almost exclusively in neuronal processes of adult tissues and is therefore an excellent marker for neurons. Neuron specific, posttranslational modifications within the C-terminal domain of beta III tubulin have been shown to be developmentally regulated suggesting that they may serve to modulate the interaction of tubulin with microtubule associated proteins (Lee et al., 1990).
