PF-562,271 is a potent, ATP-competitive, reversible inhibitor of FAK and Pyk2 catalytic activity with a IC(50) of 1.5 and 14nM, respectively. In addition, PF-562,271 displayed robust inhibition in an inducible cell-based assay measuring phospho-FAK with an IC(50) of 5nM. PF-562,271 was evaluated against multiple kinases and displays <gt/>100x selectivity against a long list of nontarget kinases. In tests, PF-562,271 inhibits FAK phosphorylation in vivo in a dose-dependent fashion with a calculated EC50 of 93ng/mL using injections.

FAIM2 Antibody: Programmed cell death regulates a number of biological processes such as normal organism development, tissue homeostasis, and removal of damaged cells. Disruption of this process has been implicated in a variety of diseases such as cancer. FAIM2 is a recently identified protein that can inhibit the apoptotic signal transduced by the Fas receptor but not from the related tumor necrosis factor-alpha death signal. In this respect, FAIM2 is functionally similar to the anti-apoptotic proteins FAIM, FLIP and Bcl-xL. FAIM2, a seven membrane spanning protein, can bind the Fas receptor but does not regulate Fas expression or inhibit binding of FADD to Fas. FAIM2 is widely distributed, but highly expressed in the hippocampus and other neural tissues. FAIM2 was also identified as the neural membrane protein 35 (NMP35) and its expression is known to be regulated by the Phosphatidylinositol 3-kinase-Akt/PKB pathway.

Potent protein kinase VEGFR2 inhibitor (2nm range). Binds to the pseudokinase domain of MLKL and blocks cell death by necroptosis, as well. With regards to its role in necroptosis, it is an ATP-mimetic that was shown to bind recombinant mouse MLKL pseudokinase domain, which showed inhibited TSQ-induced death of mouse dermal fibroblasts by delaying MLKL translocation to the membrane. Appears therefore to be a valuable reagent to inhibit necroptosis. It also provides an important proof-of-principle that targeting catalytically-dead pseudoenzymes represents a feasible, emerging therapeutic avenue.
